Neuronal NCX1 overexpression induces stroke resistance while knockout induces vulnerability via Akt

Abstract: Three different Na þ /Ca 2þ exchanger (NCX) isoforms, NCX1, NCX2, and NCX3, are expressed in brain where they play a relevant role in maintaining Na þ and Ca 2þ homeostasis. Although the neuroprotective roles of NCX2 and NCX3 in stroke have been elucidated, the relevance of NCX1 is still unknown because of embryonic lethality of its knockingout, heart dysfunctions when it is overexpressed, and the lack of selectivity in currently available drugs. To overcome these limitations we generated two conditional genet… Show more

“…Indeed, the increased NCX3 expression induced by PC is partially prevented in ncx3+/− mice, this is sufficient to prevent PC-induced protection. The fact that NCX3 may represent one of the effector of ALS PC is in line with previous works demonstrating that the genetic ablation of this transporter worsens the course of several neurological disorders such as brain ischemia, multiple sclerosis, Alzheimer disease and epilepsy 12 , 13 .…”

“…Protein content was determined by the Bradford method 38 . Percoll-purified synaptosomes were resuspended in medium B (1 mg/ml) and loaded with the ratiometric fluorescent Ca 2+ indicator Fura-2 AM (6 µM) 12 in the presence of 16 µM bovine serum albumin for 45 min at 37 °C. Dye-loaded synaptosomes were then washed by centrifugation, resuspended in medium B containing 1.2 mM CaCl 2 , and attached to poly- d -lysine-coated coverslips for 20 min at 37 °C.…”

“…An investigator blinded to group and treatment tested the mice on days 1, 7, 14, 21, and 28 after surgery. The mNSS measures motor function, gait, sensory function, balance, and reflex in a series of tests, each with its own scoring range[51]. Mice are given a score of 0 for normal function and a maximum score for no response or inability to perform the task.…”

“…This effect on nucleoplasmic Ca 2+ was due to the selective localization of XIP-NLS in the nucleus where it binded to the f-loop of nuNCX1, as also demonstrated by the coimmunoprecipitation between the peptide and nuNCX1 in isolated nuclei. Accordingly, the same interaction occurs at plasma membrane NCX level 22 . Interestingly, XIP-NLS inhibited nuNCX1 Ca 2+ n -extruding function without affecting the activity of plasma membrane NCX1, as demonstrated by patch-clamp experiments in whole neuronal cells.…”

“…In addition, outer and inner membranes of NE are provided with K + , Cl − , and R-type Ca 2+ channels 13 15 , Na + /H + exchanger 1 16 , Ca 2+ -ATPase pump 17 , and NCX1 18 , 19 . Despite the existent knowledge on the role of plasma membrane NCX in the regulation of intracellular Ca 2+ concentrations and Ca 2+ -dependent pathways 20 22 , nuNCX1 function remains unexplored. Interestingly, global silencing of NCX1 prevents neurite outgrowth through the inhibition of Akt pathway thus reducing NeuN-positive neurons 23 .…”
