Neuronal Signaling by Thy-1 in Nanodomains With Specific Ganglioside Composition: Shall We Open the Door to a New Complexity?

Abstract: Thy-1 is a small membrane glycoprotein and member of the immunoglobulin superfamily of cell adhesion molecules. It is abundantly expressed in many cell types including neurons and is anchored to the outer membrane leaflet via a glycosyl phosphatidylinositol tail. Thy-1 displays a number of interesting properties such as fast lateral diffusion, which allows it to get in and out of membrane nanodomains with different lipid composition. Thy-1 displays a broad expression in different cell types and plays confirmed… Show more

“…LRs were isolated using discontinuous sucrose density centrifugation, by an optimized procedure modified in our Laboratory (Ilic et al., 2019; Puljko et al., 2021). Frozen cortical brain tissue samples were thawed on ice and 70 ± 5 mg of tissue was homogenized in 3 ml of detergent‐free homogenization buffer (50 mM Tris (pH = 8), 150 mM NaCl, 1 mM MgCl 2 , 1 mM CaCl 2 , and protease inhibitors 1 mM phenylmethylsulfonyl fluoride (PMSF), 5 mM NaF, 1 mM Na 3 VO 4 , and 1% protease inhibitor cocktail) containing 0.32 M sucrose with 30 strokes in a Potter‐Elvehjem glass homogenizer with a Teflon pestle.…”

“…This dynamic process requires equally dynamic changes of the interactions taking place between Thy-1/CD90 and its binding partners. Thus, the lipid-protein composition of the microdomains where Thy-1/CD90 resides, and which are known to undergo constant, rapid, and dynamic organization [reviewed in Ilic et al (2019)] are ideal platforms to allow the fluid and active arrangements of their components.…”

“…Recent reports have indicated that the interaction of Thy-1/CD90 with integrin mediates association of many different cell types [reviewed in Kong et al (2013), Furlong et al (2018), Hagood (2019), Hu and Barker (2019), Ilic et al (2019), Morris (2018), and Picke et al (2018b)]. For example, Thy-1/CD90 plays an important role as a β 3 integrin ligand in fibroblasts and cancer cells.…”

“…Ganglioside head groups, protruding toward the extracellular space, significantly contribute to the cell glycocalyx and are major determinants of the features of the cell surface. In particular, they modulate complex interactions with other molecules sitting on the same cellular membrane ( cis interactions) or soluble molecules present in the extracellular environment or with molecules associated with the surface of other cells ( trans interactions) (reviewed in [2,23]). It is thus conceivable that the effects of A2B5 gangliosides are exerted by modulating the activity of receptors and their following signaling cascades [24].…”
