Neutral liposomes containing crown ether-lipids as potential DNA vectors

Abstract: Three crown ether derivatives, 1,2-O-dioleoyl-3-O-{2-[(12-crown-4)methoxy]ethyl}-sn-glycerol (12C4L), 1,2-O-dioleoyl-3-O-{2-[(15-crown-5)methoxy]ethyl}-sn-glycerol (15C5L) and 2,3-naphtho-15-crown-5 (NAP5), have been incorporated into 1-palmitoyl-2-oleoyl-phosphatydilcholine (POPC) liposomes. The size of the crown ether and the lipophilic moiety of 12C4L, 15C5L and NAP5 influence the stability and the properties of the extruded POPC liposomes determined at 25°C in buffered aqueous solution at pH7.4. The invest… Show more

“…CSCs and VM are involved in cell plasticity, which is the capability of an aberrant population to ECs transdifferentiation (Figure 1) (92). VEGF receptors regulate expression of specific marker for ECs, such as VE-Cadherin (93).…”

“…[ 28 ] synthesized and characterized a series of cholesterol and amino acid-based cationic lipids through ester and ether linkages. It has been noticed that both ether- and amide-linked lipids are more stable than the carbamate- or ester-linked lipids and provided an improved gene transfer efficiency [ 29 , 30 , 31 , 32 , 33 ]. The linker of amide and ether bonds to cationic lipids might be attributed to a stable and highly efficient gene transfer system.…”

“…In this area of interest, the self-assembled association of neutral liposomes (L), DNA and divalent metal cations (M 2+ ) in ternary L-DNA-M 2+ complexes able to transfect genetic materials into cells have been explored; [3][4][5][6][7] in this system the interaction between neutral lipids and DNA is mediated by metal cations that provide stability to the complexes. In order to improve the ability of neutral liposomes to complex DNA, our strategy was to develop liposomal gene delivery systems containing new synthetic lipids lacking in positive charge but capable of acting as effective cationic lipids.…”
