New acute myeloid leukemia-derived cell line: MUTZ-8 with 5q−

Abstract: The advent of continuous human leukemia-lymphoma cell lines as a rich resource of abundant, accessible and manipulable living cells has contributed significantly to a better understanding of the pathophysiology of hematopoietic tumors. We describe the establishment of the new continuous leukemia cell line MUTZ-8 from a patient with acute myeloid leukemia (AML): MUTZ-8 was derived from the peripheral blood of a 63-year-old woman with AML M4 (25 years after onset of myelodysplastic syndromes, MDS). DNA fingerpri… Show more

“…Cell lines were tested for JAK2 V617F expression applying the PCR-based ARMS system, a method that specifically amplifies the normal and mutant sequences plus a positive control band in a single reaction. 5 According to ARMS and direct sequencing of genomic PCR products, 5/79 cell lines carried the JAK2 V617F mutation (Figure 1a and b): MB-02 derived from a patient with a history of IM and myeloid metaplasia that developed into AML M7; 14 MUTZ-8 established from a patient with AML M4 25 years after the onset of MDS; 18 SET-2 from a patient at the stage JAK2 V617F mutation in cell lines H Quentmeier et al of leukemic transformation of ET; 19 and UKE-1 from a patient with ET, which transformed into acute leukemia. 20 The AML M6-derived cell line HEL was the only JAK2 V617F positive cell line tested lacking a known patient-history of MPD or MDS.…”

“…Kasumi-1 (the M2 subtype of AML with t(8;21) translocation, DSMZ: ACC 220), KG-1 (leukemic stem-like cells established from the bone marrow cells of an AML patient, characterization of 98% CD34 + cells, DSMZ: ACC 14), MUTZ-8 (leukemic stem-like cells, characterization of 84% CD34 + cells, ACC 689) ( Hu et al, 2002 ), NB4 (M3 subtype of AML, APL cell line, DSMZ: ACC 207), and HL-60 (the M2 subtype of AML, ACC 3) cell lines were used. Cord blood samples from three healthy individuals (obtained from the Affiliated Hospital of Weifang Medical University, Weifang, China) were collected after obtaining written informed consent from the donors.…”
