New Diarylmethylpiperazines as Potent and Selective Nonpeptidic δ Opioid Receptor Agonists with Increased In Vitro Metabolic Stability

Abstract: Nonpeptide delta opioid agonists are analgesics with a potentially improved side-effect and abuse liability profile, compared to classical opioids. Andrews analysis of the NIH nonpeptide lead SNC-80 suggested the removal of substituents not predicted to contribute to binding. This approach led to a simplified lead, N, N-diethyl-4-[phenyl(1-piperazinyl)methyl]benzamide (1), which retained potent binding affinity and selectivity to the human delta receptor (IC(50) = 11 nM, mu/delta = 740, kappa/delta > 900) and … Show more

“…Data are presented as means Ϯ SEM. sections adjacent to those used for in situ hybridization were processed for receptor autoradiography using [ 125 I]-deltorphin-II ([ 125 I]-DLT-II) (Plobeck et al, 2000). The peptide (BACHEM Bioscience, King of Prussia, PA) was iodinated according to the chloramine T method as described previously (Fraser et al, 1999) (specific activity, 2200 Ci/mmol).…”

“…125 I]-DELT-II (Plobeck et al, 2000; iodination was performed using the chloramine T method; specific activity was considered to be 2000 Ci/mmol, the theoretical value). Sections were preincubated for 30 minutes at room temperature in 50 mM Tris-HCl, 120 mM NaCl, and 1 mM MgCl 2 buffer (pH 7.4) and then incubated for 60 minutes at room temperature in the same buffer containing 0.5% bovine serum albumin (BSA), 0.1 mM Bestatin, 0.1 mM phenylmethylsulfonyl fluoride (PMSF), and 40 pM […”

“…This GPCR protein is located in the central nervous system and to a lesser extent in peripheral tissues [19]. CB-1 antagonists and inverse agonists have been reported to influence memory as well as appetite control and smoking cessation which makes this receptor an attractive drug target for therapeutic intervention [11].…”
