New traits in crops produced by genome editing techniques based on deletions

Abstract: One of the most promising New Plant Breeding Techniques is genome editing (also called gene editing) with the help of a programmable site-directed nuclease (SDN). In this review, we focus on SDN-1, which is the generation of small deletions or insertions (indels) at a precisely defined location in the genome with zinc finger nucleases (ZFN), TALENs, or CRISPR-Cas9. The programmable nuclease is used to induce a double-strand break in the DNA, while the repair is left to the plant cell itself, and mistakes are i… Show more

“…Furthermore, gene editing has shown great promise in creating new allelic variants, using various technologies such zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and clustered regularly interspaced short palindromic repeats (CRISPR) [145][146][147][148]. Gene editing can result in gene modification (e.g.…”

“…However, gene editing approaches are still in their infancy, and further improvements are necessary. For instance, despite its high specificity, the CRISPR-Cas9 method may still have potential off-target effects, which could be reduced by increasing the specificity of Cas9, the RNA-guided DNA endonuclease protein, and the guide RNA design, or using alternative endonucleases with higher specificity [147]. One of the steps in gene editing that also needs to be refined is the delivery of the new genetic material.…”

“…However, the same methods can also be used to silence endogenous genes. This is especially important in crop improvement, because many advantageous traits, such as loss of shattering, are often caused by recessive loss-of-function mutations [147]. So far, most of these recent gene editing techniques have focused on disease resistance in crops, but same techniques can be used to improve other traits including shattering-resistance, reduced allergenicity, abiotic stress tolerance, and increased nutritional value [141].…”

“…Site-directed nucleases induce double-strand breaks (DSBs) in the DNA which are subsequently repaired by the hosts own cellular mechanisms. The type of repair can be categorized in three main types [11][12][13][14]:…”
