Nicotinic Acetylcholine Receptor Subtypes Expression during Rat Retina Development and Their Regulation by Visual Experience

Abstract: By acting through retinal nicotinic acetylcholine receptors (nAChRs), acetylcholine plays an important role in the development of both the retina and central visual pathways. Ligand binding and immunoprecipitation studies with subunit-specific antibodies showed that the expression of ␣Bungarotoxin (␣Bgtx) and high-affinity epibatidine (Epi) receptors is regulated developmentally and increases until postnatal day 21 (P21). The increase in Epi receptors is caused by a selective increase in the subtypes containin… Show more

“…In mouse and rat CNS, a6-and b3-subunits colocalize in dopaminergic neurons and retina 62,63 as a6b2b3* or a4a6b2b3 nAChRs. [64][65][66] a6 nAChRs are expressed in higher concentration on nerve terminals than on cell bodies or dendrites, suggesting that a6 or another subunit (such as b3) plays a targeting role.…”

“…In situ hybrydisation studies have shown that the α2β2* subtype is highly expressed in many regions of primate brain [61,62], whereas its expression in rodents is limited to retina and interpeduncular nucleus (IPn) [63,64].…”

“…6OHDA caused an almost complete loss of DA cell structures as shown by a Ͼ90% decrease in [ 3 H]WIN 35,428 binding (data not shown). Since the retina is a major source of nAChR subtypes with uncommon neuronal subunits (such as ␣2, ␣3, ␣5, ␣6, and ␤3) (Moretti et al, 2004;Gotti et al, 2005b;Cox et al, 2008), we performed bilateral eye enucleation to assess a possible contribution of these minor subtypes from visual afferents. A third major source of ␣6* nAChRs in the brain is the locus ceruleus, where high levels of ␣6 mRNA have been detected (Le Novère et al, 1996).…”
