“…To clarify whether iRhom2 regulates TNF-α secretion in macrophages, RAW264.7 cells were transfected with iRhom2 small interfering RNA (siRNA) before LPS stimulation, and TNF-α secretion by LPS-stimulated RAW264.7 cells was measured by enzyme-linked immunosorbent assay (ELISA). Before LPS stimulation, RAW264.7 cells were transfected with iRhom2 or control siRNA using Lipofectamine RNAiMax (Invitrogen) 17 , 18 , 20 . Twenty-four hours after transfection, the cells were stimulated with LPS (1 µg/ml LPS for 4 h), and the concentration of TNF-α in the culture supernatants was measured using a commercially available ELISA kit (R&D Systems, Minneapolis, MN, USA).…”

“…Nimbolide is an important phytochemical isolated from the tree, which considerably reduced the expression of inflammatory cytokines interleukin (IL‐6, IL‐8, and IL‐12,) and nuclear factor (TNF‐α) in intestinal epithelial cell line. Nimbolide inhibited NF‐κB signaling in intestinal epithelial cells and macrophages and improved experimental colitis in mice (Seo et al, ).…”

“…Use of Wasabia japonica , a plant abundant in phytochemicals, in DSS-induced colitis inhibited the activation of NF-κB, the secretion of pro-inflammatory cytokines and prevented the onset of colitis at high dosages [ 27 ]. Similarly, Nimbolide, a phytochemical isolated from the Neem ( Azadirachta indica ) tree, alleviated both acute and chronic colitis induced in mice and inhibited NF-κB activation in macrophages [ 28 ].…”
