NKT and CD8 lymphocytes mediate suppression of hepatocellular carcinoma growth via tumor antigen‐pulsed dendritic cells

Abstract: Dendritic cells (DCs) are antigen presenting cells that play a role in T-cell activation. Liver-associated natural killer T lymphocytes (NKTs) are a unique subset of lymphocytes that may be important in antitumor immunity. Hepatitis B virus (HBV)-associated hepatocellular carcinoma (HCC) expresses hepatitis B virus surface antigen (HBsAg) on its cell surface and may serve as a tumor-associated antigen. The aim of the study was to evaluate the antitumor effect of DC pulsed with tumor or viral-associated antigen… Show more

“…Tumor antigen-pulsed dendritic cells effectively suppressed the growth of HCCs in mice and this effect was associated with enhanced CD8+ lymphocyte function [8]. A previous study by the authors demonstrated a significant positive correlation between the CD8+ T cell density of tumors and the occurrence of spontaneous apoptosis in cancer cells [1].…”

“…5,6 NKT-like cells have been shown to mediate suppression of hepatocellular carcinoma following the adoptive transfer of tumor lines. 98,99 Production of IFN-␥ and IL-12 by NKT cells and dendritic cells, respectively, appears to be responsible for the antitumor effect against either primary or metastatic disease in these models, 13,97,100,101 although activated invariant NKT cells can have NK/lymphokine-activated killing-like and/or CD1d-specific cytoxic activity, 11,12,14,55 which could in principle contribute to antitumor responses.…”

“…In a murine model of hepatocellular carcinoma (HCC), NKT cells were shown to play a role in oral immune regulation using HCC lysates and hepatitis B virus (HBV) envelope proteins, as well as a role in the adoptive transfer of DCs pulsed ex vivo with these antigens [Margalit et al 2005;Shibolet et al 2003]. …”
