Non-coding RNA in cancer

Abstract: Majority of the human genome is transcribed to RNAs that do not encode proteins. These non-coding RNAs (ncRNAs) play crucial roles in regulating the initiation and progression of various cancers. Given the importance of the ncRNAs, the roles of ncRNAs in cancers have been reviewed elsewhere. Thus, in this review, we mainly focus on the recent studies of the function, regulatory mechanism and therapeutic potential of the ncRNAs including microRNA (miRNA), long ncRNA (lncRNA), circular RNA (circRNA) and PIWI int… Show more

“…The entire mammalian genome can be transcribed to some level, yet there remain thousands of RNA transcripts that do not code for proteins. These are known as non-coding RNAs (ncRNAs) ( Palazzo and Lee, 2015 ; Yan and Bu, 2021 ; Yang et al, 2020 ; van Bakel et al, 2010 ). Their expression level and the function are very controversial.…”

“…Based on their size, shape and genomic location, ncRNAs are divided into three major classes, long ncRNAs (lncRNAs), circular RNAs (circRNAs) and short ncRNAs (micro RNAs, short interfering RNAs, piwi RNAs) ( Palazzo and Lee, 2015 ; Wu et al, 2019 ; Yan and Bu, 2021 ). Long ncRNAs (lncRNA) are the linear gene transcripts with limited protein-coding capacities, over 200 NT long and regulate gene expression at pre-transcriptional, transcriptional, and post-transcriptional levels ( Yan and Bu, 2021 ). Depending on their expression pattern and biological function, lncRNAs can be classified as sense, antisense, bidirectional, intron, intergenic, or enhancer-lncRNAs ( Al-Rugeebah et al, 2019 ; Palazzo and Lee, 2015 ).…”

“…Most circRNAs generated by reverse splicing are found mainly in the cytoplasm, and translation of linear mRNAs usually requires the structure of a 5′ end cap and a 3′ end poly(A) tail to remain stable. CircRNAs are not normally considered to have a translational function ( Marquez-Molins et al, 2021 ; Yan and Bu, 2021 ) but they can initiate translation in the cell if they have an internal ribosome entry site (IRES) ( He L. et al, 2021 ; Wang et al, 2021b ; Sinha et al, 2021 ). This approach provides more polypeptide sequences and increases the polypeptide yield because the ribosome does not need to bind to RNA template repeats ( Li P. et al, 2021 ).…”

“…Recent findings have highlighted that noncoding RNAs expression is systematically altered in cancers (Vo et al, 2019; Yan & Bu, 2021), interpreting the essential role of noncoding RNAs on tumorigenesis, growth, invasion, and progression. Among them, miRNA, lncRNA, and circRNA are the three major ncRNA subtypes with distinct functions in Human malignancies.…”
