Non-thermal, pulsed electric field cell ablation: A novel tool for regenerative medicine and scarless skin regeneration

Abstract: High voltage, short pulsed electric fields (PEF) is a non-thermal ablation method, in which defined PEF irreversibly destabilize cell membranes, while preserving other tissue components such as the extracellular matrix (ECM). In the present report, we show that PEF ablated rat skin retains its microvascular blood supply and ECM structure. Complete regeneration of epidermis, hair follicles, sebaceous glands, and the panniculus carnosusis observed two months after the ablation. Our results clearly indicate that … Show more

“…In NTIRE, externally applied pulsed electric fields cause irreversible damage to cells by affecting the cell membrane, while sparing the tissue scaffold, large blood vessels, and other tissue structures (83)(84)(85). The preservation of tissue architecture is an important and unique property of NTIRE ablation, and it probably contributes to the reduced scarring observed (as reported in 83,84,86).…”

“…To address the effects of PEF on nontarget tissue, we previously investigated the healing process of normal rat skin ablated by PEF. 38 In this study, we showed that PEF-ablated normal skin regenerated rapidly without scars. 38 Our results show that PEFs preserve the extracellular matrix and the microvasculature of the ablated area—properties that most likely promote scarless skin regeneration.…”

“…These limitations include: a significant impact of all factors (listed in Section 1 -such as undisclosed diseases, hot meals consumed before the test, lack of acclimatisation to the room etc. [44][45][46][47][48][49][50]) -on temperature distribution. All these factors can fully disqualify the measurement.…”
