Nondisjunction and unequal spindle organization accompany the drive of Aegilops speltoides B chromosomes

Abstract: Summary B chromosomes (Bs) are supernumerary chromosomes, which are often preferentially inherited. When transmission rates of chromosomes are higher than 0.5, not obeying the Mendelian law of equal segregation, the resulting transmission advantage is collectively referred to as ‘chromosome drive’. Here we analysed the drive mechanism of Aegilops speltoides Bs. The repeat AesTR‐183 of A. speltoides Bs, which also can be detected on the Bs of Aegilops mutica and rye, was used to track Bs during pollen develop… Show more

“…Mature ovules were fixed in 1× phosphate-buffered saline (PBS) containing 4% paraformaldehyde (PFA) under vacuum at 4 °C for 10 min followed by a 5 h fixation at 4 °C without vacuum. Fixed cowpea ovules were embedded and sectioned according to (Wu et al 2019). For the analysis of pollen mother cells, immature anthers were fixed with 1× PBS containing 4% PFA under vacuum at 4 °C for 10 min followed by a 30 min fixation at 4 °C without vacuum.…”

“…At the beginning of the tillering stage, plants were kept for one month at 12 °C to ensure synchronous flowering. Presence of Bs in plants was identified by PCR with primers for the B-specific repeat AesTR-183 [ 26 ] ( Table S1, Supplementary Materials ) and the exact number of Bs was determined by flow-cytometric analysis according to [ 27 ]. Only plants with 2, 3 or 4 Bs were used for the generation of embryos.…”

“…In Ae. speltoides , for instance, Bs accumulate in the generative nuclei to more than 93% [Wu et al, 2019]. In order to decipher the molecular biology behind the process of B chromosome drive, Anastassia Boudichevskaia (Gatersleben, Germany) demonstrated how comparative RNAseq analysis was used to identify rye Bdrive-associated transcripts.…”
