Normalized enzyme-linked immunosorbent assay for determining immunoglobulin G antibodies to cytomegalovirus

Abstract: A normalized enzyme-linked immunosorbent assay for the determination of immunoglobulin G antibodies to cytomegalovirus is described. The rapid assay involves three 30-min incubations and permits the quantitation of antibody levels with a single-specimen dilution in conjunction with a reference antibody preparation. The results obtained with the normalized enzyme-linked immunosorbent assay correlated closely with the results of complement fixation titrations and another commercially available enzyme-linked immu… Show more

“…These findings imply that the antibody responses to these infections are independent of each other in AIDS patients. It also supports the lack of cross-reactivity between the antigens used, in confirmation of previous absorption studies (6).…”

“…The amount of control antigen used reduced the specific absorbance by about 20% but improved the discrimination between positive and negative reactions. The false-positive reactions with the CMV conjugate could have been due to the presence of antinuclear antibodies (2,10,13), which react with enzyme-labeled nuclear antigens in the CMV conjugate.…”

“…We found a Spearman's correlation coefficient (rs) of 0.79 (P < 0.001). Kiefer et al reported a correlation coefficient (r) of 0.67 when testing 121 CF-positive serum samples by an indirect IgG ELISA (10). The considerable amount of scatter observed among the quantitative ELISA values at each CF titer suggested that the two methods measure different antibodies.…”

“…Numerous diagnostic modalities have been applied to the detection of CMV (Doerr et al, 1985). Routine serologic analysis of circulating anti-CMV antibodies by radioimmunoassay (RIA) or enzyme-linked immunosorbant assay (ELISA) (Griffiths, 1985;Kiefer et al, 1983;McKeating et al, 1985;Nath et al, 1987) is of little value in AIDS patients. Immunosuppressed patients frequently fail to mount a serologically detectable antibody response to CMV (Doerr et al, 1977).…”
