Novel Fluorometric Assay for Hydroxyl Radical Scavenging Capacity (HOSC) Estimation

Abstract: A novel fluorometric method was developed and validated for hydroxyl radical scavenging capacity (HOSC) estimation using fluorescein as the probe. A constant flux of pure hydroxyl radical is generated under physiological pH using a Fenton-like Fe3+/H2O2 reaction. The generation of pure hydroxyl radicals under the experimental conditions was evaluated and confirmed using electron spin resonance with DMPO spin-trapping measurements. The hydroxyl radical scavenging capacity of a selected antioxidant sample is qua… Show more

“…Therefore, · OH was suggested to be an excellent target to investigate antioxidants in the fields of food, nutrition, health, and medicines. 21,22) In addition, ESR is widely used as a prerequisite tool to study free radicals even with suspensions or pastes, 23) and is thought to be useful to study slightly insoluble less-polar ginsenosides or MRPs. Especially, some triterpenoid glycosides and MRPs are known to have transition metal chelating and reducing capabilities closely related to · OH scavenging activity.…”

“…Hydroxyl radical scavenging capacities were investigated using a Victor 3 multilabel plate reader (PerkinElmer) according to a laboratory protocol described earlier [12]. Hydroxyl radicals were generated by a Fenton-like reaction using H 2 O 2 and Fe 3+ , and trolox was used to prepare the standard curve.…”

“…For example, Moore et al 43 noticed that randomly-methylated β-cyclodextrin could reduce the sensitivity of hydroxyl radical scavenging capacity (HOSC) measurements, since their experimental findings indicated the possibility of the • OH quenching of Me-β-CD. 43 García-Padial et al 44 pointed out that cyclodextrins may yield high blank values in the ORAC-FL antioxidant activity assay, and that intermolecular H-bonding between the cyclodextrin and the antioxidant it includes may attenuate the measured antioxidant activity. 44 It was found in this work that the complexed BC with β-CDs was a less effective antioxidant than free BC, considering the TEAC value of β-carotene in acetonated medium previously given as 3.10.…”
