Novel Luciferase Reporter System for In Vitro and Organ-Specific Monitoring of Differential Gene Expression inListeria monocytogenes

Abstract: In this paper we describe construction of a luciferase-based vector, pPL2lux, and use of this vector to study gene expression in Listeria monocytogenes. pPL2lux is a derivative of the listerial integration vector pPL2 and harbors a synthetic luxABCDE operon encoding a fatty acid reductase complex (LuxCDE) involved in synthesis of the fatty aldehyde substrate for the bioluminescence reaction catalyzed by the LuxAB luciferase. We constructed pPL2lux derivatives in which the secA and hlyA promoters were translati… Show more

“…For example, strains of E. coli rendered metabolically dependent on diaminopimelic acid (DPI) have been generated, permitting maintenance of plasmids featuring DPI-producing genes as plasmid loss lead to bacterial cell death. 68,69 Transposon, 46,70 transducing bacteriophages, 42 temperature sensitive vectors 71 and bacteriophage integrase systems [72][73][74] have been developed to integrate genes into the chromosomes of selected bacteria. However, it is important to stress that the maintenance and expression of high levels of recombinant DNA may place an unwelcome metabolic burden microorganisms.…”

“…The promoter of llsA was amplified by using oligos PllsAmonoFlong and PllsAmonInbluntR (Table S2), digested with XhoI, and cloned into SwaISalI-digested pPL2 lux as described (29). The resultant plasmid pPL2 llsA:lux was isolated from E. coli and introduced into L. monocytogenes F2365 generating F2365 llsA:lux .…”

“…salivarius UCC118 Reduces Listeria Infection in Mice. Recently, a luciferase-based reporter system was developed to track systemic Listeria infection in the murine model (14). We used a derivative of this system, in which the hly promoter was translationally fused to luxABCDE and integrated into the chromosome of L. monocytogenes EGDe.…”
