Novel Multitarget Real-Time PCR Assay for Rapid Detection of Bordetella Species in Clinical Specimens

Abstract: A novel multitarget real-time PCR (RT-PCR) assay for the rapid identification of Bordetella pertussis, B. parapertussis, and B. holmesii was developed using multicopy insertion sequences (ISs) in combination with the pertussis toxin subunit S1 (ptxS1) singleplex assay. The RT-PCR targets for the multiplex assay include IS481, commonly found in B. pertussis and B. holmesii; IS1001 of B. parapertussis; and the IS1001-like sequence of B. holmesii. “…This sequence is often the target of choice because it is found in multiple copies in the genome of B. pertussis (50-238 copies per genome), making this test much more sensitive. However, because of its high copy number, false positive results may be generated and positive results using only a target may lead to a false diagnosis of pertussis (15,22,23). The IS481 is also present in B. holmesii (8-10 copies per genome) and in B. Bronchiseptica, but the presence of IS481 is host dependent in this specie and is found in several animals, being rare human isolates (24).…”
