Novel Nanocarriers for Targeted Topical Skin Delivery of the Antioxidant Resveratrol

Abstract: Resveratrol (RSV) is a potent lipophilic antioxidant with a low aqueous solubility. Novel nanoformulations have been successfully developed and evaluated to increase the potential of resveratrol as a skin targeting antioxidant. Nanoformulations were prepared using a spontaneous emulsification method, and characterized and evaluated for their capabilities to penetrate/permeate the skin. In nanoformulations, the thermodynamic activity of the RSV penetration into/permeation through the skin was correlated with th… Show more

“…However, one can see that thanks to coumarin encapsulation in the nanoemulsion, the latter penetrates the skin (examined 1 h after application), stopping at the dermis border ( Figure 5 C,D). Similar results were obtained by Sarheed et al [ 59 ], Yousef et al [ 36 ] and Nastiti et al [ 60 ], whereby the encapsulation of loratadine, curcumin and resveratrol, respectively, in a nanoemulsion enhanced the permeability of these compounds through the skin.…”

“…Such high doses result in adverse effects (e.g., diarrhea, nausea, and abdominal pain with >1 g/day) [402]. As the poor bioavailability limits the RSV activity, there are various approaches for overwhelming the bioavailability, including co-delivery with piperine prodrug [403], micronized powders [403], and nanoplatform delivery [404][405][406][407]. Application of nanomedicine can improve the stability and bioavailability, and minimize side effects of RSV, which is making RSV a prospective candidate for treating many diseases, including cancers.…”

“…Recently, various approaches have been proposed to improve RSV biological activities after systemic and topical administration; these strategies include RSV incorporation into different types of nanocarriers and synthesis of prodrugs or RSV derivatives with improved chemical–physical characteristics compared to the parent drug [ 15 , 16 , 17 , 18 , 19 , 20 , 21 , 22 , 23 ]. In particular, several studies suggest the feasibility of improving RSV effectiveness by synthesizing derivatives in which molecules that might exert a synergic effect are linked to RSV (i.e., RSV hybrids and codrugs) [ 24 , 25 ].…”
