Novel Parallelized Quadrupole/Linear Ion Trap/Orbitrap Tribrid Mass Spectrometer Improving Proteome Coverage and Peptide Identification Rates

Abstract: Proteome coverage and peptide identification rates have historically advanced in line with improvements to the detection limits and acquisition rate of the mass spectrometer. For a linear ion trap/Orbitrap hybrid, the acquisition rate has been limited primarily by the duration of the ion accumulation and analysis steps. It is shown here that the spectral acquisition rate can be significantly improved through extensive parallelization of the acquisition process using a novel mass spectrometer incorporating quad… Show more

“…Orbitrap Tribrid HRMS combines the technologies of quadrupole, linear ion trap and orbitrap. The separation of ion isolation, fragmentation and detection could be achieved by Tribrid Orbitrap spatially and temporally compared with the other orbitrap instruments with increased data acquisition rates [27]. To our knowledge, similar application of Tribrid Orbitrap instrument on the simultaneous qualitative and quantitative analysis of polyfluoroalkyl substances has not been conducted.…”

“…Similar to FTICR, the frequency domain is converted to the m/z domain using a calibration equation [82]. Continual improvements in Orbitrap MS instrumentation have resulted in the commercialization of numerous configurations of the Orbitrap mass analyzer, such as the LTQOrbitrap MS [129], the quadrupole-Orbitrap MS (QExactive) [130], and the quadrupole -LTQOrbitrap MS (Orbitrap Fusion) by Thermo Fisher Scientific [131]. A particular advantage of Orbitrap MS over FTICR MS in the context of top-down analysis involves the relationship between m/z and resolving power.…”

“…The ETD hardware is adapted from the Thermo Fusion [23] and follows from the work of Earley et al [24] Fluoranthene reagent ions are generated by Townsend discharge in an excess of nitrogen gas, in an ionization volume located between the slens and the first quadrupole ion guide in the Velos Pro (termed Bfront-ETD^or BFETD^by the inventors). The analyte and reagent targets are 200,000 charges with a reaction period of 10 ms for dissociation of the 34+ charge state of carbonic anhydrase in the Velos Pro linear trap.…”
