Nuclear Factor-erythroid 2 (NF-E2) p45-related Factor-2 (Nrf2) Modulates Dendritic Cell Immune Function through Regulation of p38 MAPK-cAMP-responsive Element Binding Protein/Activating Transcription Factor 1 Signaling

Abstract: Background: Nrf2 is required for normal dendritic cell immune functions. Results: Loss of Nrf2 alters DC function and results in hyperphosphorylation of CREB/ATF1 transcription factors that are responsive to p38 MAPK inhibition. Conclusion: The p38 MAPK-CREB/ATF1 axis contributes to Nrf2-mediated regulation of DC function.Significance: Defining the relevance of p38-CREB/ATF1 in Nrf2 signaling expands understanding of DC biology.

“…Previous studies have shown that ROS can activate the MAPK, NF-kB, and PI3K/Akt pathways in DCs (8,35). Our work showed that treatment with PLM or NAC (Y. Xiao, et al, unpublished data) blocked LPS-induced phosphorylation of p38 and JNK, as well as NF-kB and PI3K/Akt pathways.…”

“…Indeed, NRF2 not only acts as master regulator of the antioxidative response but was likewise shown to negatively control DC maturation (25)(26)(27)(28).…”

“…There is also increasing evidence that NRF2 may control the differentiation and function of dendritic cells (DCs) and macrophages involved in antigen presentation and regulation of adaptive immune responses. In fact, NRF2 deficiency was shown to alter the function and phenotype of DCs by increasing the expression of costimulatory molecules and consequently the antigenspecific T cell reactivity (Al-Huseini et al, 2013).…”
