Nucleoside Diphosphate Kinase Escalates A-to-C Mutations in MutT-Deficient Strains of Escherichia coli

Abstract: Nucleoside diphosphate kinase (NDK), a ubiquitous enzyme, is known for its critical role in homeostasis of cellular nucleotide pools. However, NDK has now emerged as a molecule with pleiotropic effects in DNA repair, protein phosphorylation, gene expression, tumor metastasis, development, and pathogen virulence and persistence inside the host. In this study, we reveal an unexpected role of NDK in genome instability because of its activity in converting 8-O-dGDP to 8-O-dGTP. This observation has important conse… Show more

“…A total of 32 Kan R mutants were collected, 25 that arose from S-P setup, and 7 from the P-P setup. To determine the mutation frequency (MF) accurately, we measured resistance to the antibiotics Rifampicin (Rif) and Kan (Fowler et al, 1994; Kapoor et al, 2019; Miller et al, 1977) in both populations. Compared to P cells, S cells of E. coli exhibited a ~5-fold higher MF (Figure 1E).…”

“…This process prevents the incorporation of abnormal nucleotides caused by oxidative stress into DNA, which can lead to DNA damage. The deletion of MutT results in a greatly increased mutation rate of AT-CG in E. coli [13]. MazG in Mycobacterium tuberculosis hydrolyzes dUTP, 2-hydroxy-dATP and 8-oxo-dGTP.…”

“…All of the enzymes and reaction components needed for GMP manufacture of mRNA are available from commercial providers as synthesized chemicals or bacterially produced, animal componentfree reagents [271]. GMP-grade traceable plasmid DNA, phage polymerases, capping enzymes, and NTPs are available [272]. Other ingredients are pricey or unavailable [272].…”
