Nucleoside, IX. Synthese und Eigenschaften von Lumazin‐nucleosiden — Strukturanaloga des Uridins und Thymidins

Abstract

Die Umsctzungen von Luniazin (l), seine, 6,7-Dimethyl-(2) und 6,7-Diphenyl-Derivat (3) init I-Hidogcn-bzw. I-Acyloxyribosc-und 2-Dcsoxyribose-Derivaten nach der Silyl-Methode werden beschrieben. Als Hauptproduktc werden die Lumarin-N-1-nucleoside gebildet. l ,?-Diglycoside entstehen in untergeordnetcm MaBe, wiihrend die N-3-Nucleoside meist nur in Spuren nachwcisbar sind. Die Strukturen dcr neu synthetisierten Produkte werden dnrch UV-und NMR-spcktroskopische Untersuchungen gcsichert. Thc main reaction products turned out to be thc lumazine-N-I nucleosides. 1,3-Diglycosides are formcd in minor amounts whereas iii most cases the N-3 nucleosides are present only in trace quantities. The structures of the newly synthesized reaction products have been established by u. v. and n. m. r. spectra.Die erfolgreiche Synthese von Pteridin-N-8-glycosiden 2), welche als Strukturanaloge der Purin-nucleoside angesehen werden konnen, war ausschlaggebend fiir analoge Untersuchungen zur Direktglycosidierung auch am Pyrimidinteil des Pteridingerustes. Das Ziel war die bevorzugte N-1-Substitution, urn so die strukturclle Verwandtschaft zu den Pyrimidin-nucleosiden herzustellen. Da die Synthesemethode in der Birkof~r-Variante3) der Hilbert-Johnson-Reaktion4) besteh t und als Voraussetzung das Vorhandensein von Amidfunktionen erfordert, fie1 unsere Wahl zunachst auf das Lurnazinsystems) als Ausgangsniaterkdl.
