Nylon-screen filtration devices for crude separation of dissociated cells for primary culture

Abstract: SUMMARY: This report describes the assembly and use of several types of filtration devices for use in the preparation of dissociated cells for primary culture. These Nitex-screen filter units are assembled from common, inexpensive laboratory materials. They provide a convenient means of filtering cell suspensions under sterile conditions and are adaptable for large and small volume work.

“…Repeat this step. Note: At this point it may be useful, depending on the specific application for the cell preparation, to filter the cell suspension through size-bruiting screen such as Nitex cloth (11). Exercise caution, because too fine a mesh may retain a substantial amount of the cell yield, particularly if polymerized DNA from lysed cells has not been diluted out.…”

“…The cells were trypsinized and then washed twice in medium containing 0.1% soybean trypsin inhibitor (Boehringer-Mannheim, Indianapolis, IN). Cells were resuspended in culture medium to give 2.5 x 105 cells/ml and then gently triturated through an 18-gauge cannula and filtered (McAteer and Cavanagh, 1983) through 20-pm Nitex screen (Tetko Inc., Elmsford, NY). The dispersed cell suspension (4.3 ml) was added to a chilled collagen-medium solution.…”
