Bisphenol A (BPA) is an emerging contaminant, regularly detected in aquatic ecosystems, considered as an endocrine disrupting compound (EDC). Caffeine is another chemical related to human activity, often found in surface waters. The objective of this study was to evaluate the ecotoxicological risk due to BPA and caffeine in water samples from the Sinos River basin, Rio Grande do Sul, Brazil. Water samples were collected at three sites monthly from May 9 th, 2016 to April 11th, 2017 (n = 36). BPA concentrations in water samples collected were in the range of not detected to 517 ng L-1 and caffeine concentrations in the range of 41.7 to 28,439.6 ng L-1. The concentration of BPA in the analyzed samples had a moderate correlation with caffeine (rs = 0.402). High ecotoxicological risk for BPA was characterized in 77.77% of samples, with 11.11% presenting medium and 11.1% presenting low risk. For caffeine 13.9%, 50% and 36.11% of the samples presented high, medium and low risk, respectively. Caffeine concentrations in water can be used as predictors of BPA concentrations above 10 ng L-1, the lower concentration of ecotoxicological risk, with specificity of 66.7% and sensitivity of 70.4%. The assessment of aquatic risks has shown that both investigated compounds pose risks to organisms in the studied surface waters, mouth of the Pampa stream, mouth of the Luiz Rau stream and catchment point for public supply in Lomba Grande.

Sewage release results in the disposal of various pollutants in water bodies, including the so-called emerging pollutants. Among these are the endocrine disrupters, exogenous agents that mimic the natural hormones binding to cell receptor sites activating or blocking cell signaling pathways that trigger hormone function. These pollutants have been found in environmental samples all over the world, including Brazil, at low concentrations causing effects to exposed organisms. In this context, the aims of this study were to investigate the occurrence of three emerging pollutants with estrogenic activity (17β-estradiol-E2, 17α-ethinylestradiol-EE2 and bisphenol A-BPA) in water samples from Paranoá Lake and effluent of Sewage Treatment Plants (STPs), North and South, in Brasília-DF; Evaluate the toxicity of both environmental samples as the pollutant most frequently detected (BPA) to zebrafish embryonic development. In addition, it was tested, in the same animal model, sub lethal doses of BPA by biochemical markers. Finally, the environmental concentrations detected were compared with effect concentrations, described in literature, for aquatic animals from the first and second trophic levels. In the quantification analysis, samples were processed by solid phase extraction (SPE) and subjected to an analytical method developed in HPLC/DAD/FL. In this case, the mobile phase was acetonitrile with water in the ratio 40:60 (v/v), acidified with trifluoroacetic acid (0,12%) in isocratic condition. All the toxicity assays was performed following the OECD Protocol 236 (Guideline on Fish Embryo Toxicity Test-FET). The results showed that BPA was the most predominant pollutant in analyzed samples, with 100% of detection in the treated wastewater and 23% in surface water. The concentrations found were between <394-1231 ng L-1 in the effluent and <197-644 ng L-1 in Lake samples. Other compounds, E2 and EE2, showed low frequency of detection (<18%) in both Lake and effluent samples and, for only one effluent sample, EE2 was quantified in concentration of 591 ng L-1 .Toxicity tests showed that water samples from Paranoá Lake and South STP effluent were no toxic to zebrafish embryos; BPA is slightly toxic to zebrafish embryos (LC50 13.45 mg L-1) and was rapidly degraded in an aqueous medium (4-5 days). However, effects on hatching (CEO = 10 mg L-1), delayed pigmentation, craniofacial formation (CEO = 21.54 mg L-1) and inhibition of GST (CEO = 11.68 ug L-1) were observed in embryos exposed to BPA. In addition, the levels at which the three pollutants were found in the analyzed samples are among the effects concentrations reported in the literature for aquatic animals of the first and second trophic levels, with the exception of BPA for primary consumers. Moreover, the literature suggests that BPA tends to be less toxic than E2 and EE2. O atual estilo de vida arquitetou-se gradualmente sobre materiais sintéticos feitos de uma ampla gama de polímeros à base de óleo, e isso contribuiu de alguma forma para o desenvolvimento global. Sob a perspectiva econômica para aprimorar as características e aplicações dos plásticos, os cientistas fizeram extensos testes com moléculas conhecidas e novas para atuar como dependências químicas, obtendo um tremendo sucesso com o bisfenol A (BPA). Ao longo dos anos, no entanto, as agências mundiais de vigilância em saúde discutiram os efeitos perigosos do BPA nos seres humanos com base em relatórios científicos disponíveis na época. Dessa forma, o presente trabalho comprovou a viabilidade do teste da mancha de cloreto férrico como detector de compostos fenólicos liberados a altas temperaturas de itens de plástico, o que serve como método indireto para identificação de BPA. Portanto, esta pesquisa é uma nova maneira de reconhecer compostos fenólicos nocivos em itens de plástico comercializados usando uma abordagem química simples e econômica.
