Oceanobacillus halophilus sp. nov., a novel moderately halophilic bacterium from a hypersaline lake

Abstract: A moderately halophilic bacterium was isolated from a brine sample of a hypersaline lake, Aran-Bidgol, in Iran. The strain, designated J8BT , was Gram-stain-positive, endospore-forming, rod-shaped, strictly aerobic, motile and produced cream colonies. Strain J8B T grew in NaCl at between 3.0-15.0 % (w/v) (optimally at 7.5 % NaCl, w/v), between pH 6.5-9.0 (optimally at pH 8.0) and between 20-45 8C (optimally at 35 8C). Phylogenetic analysis, based on 16S rRNA gene sequences, revealed that strain J8B T is a memb… Show more

“…Oceanobacillus, which was represented by O. profundus (8.79%-9.84%) and a minor fraction of unclassified Oceanobacillus spp. The Oceanobacillus genus consists of Gram-positive, endospore-forming, and moderately halophilic bacteria, which are frequently isolated from hypersaline lakes, salty lakes, marine sediments and seawater (Amoozegar et al 2016).…”

“…The physiology of the moderate and extreme halophilic bacteria is affected by changes in the salt concentration, growth temperature, pH and nature of available nutrients (Amoozegar et al 2016). Moderate halophilic bacteria can grow at 0.85 to 3.4M NaCl concentrations (Oren 2012; DasSarma and DasSarma 2015).…”

“…The core genes (defined as their encoded proteins having >0.4 identity to their counterparts in members of the genus and selected using CD-HIT software v4.8.1 [44]) of our isolates with available genomes of their corresponding genus and the respective outgroup were used to reconstruct the NJ phylogenomic trees, which were built using FastTree v2.1 [45] and visualized using Dendroscope v3.5.9 [46]. Outgroups were chosen and used to root the trees as follows: Bifidobacterium bifidum ATCC 29521 T for strains 299 T and 340 [10], Rhodococcus equi DSM20307 T for strains 2184 T , 2185, 2183 T and 2189 [47], and Salimicrobium flavidum DSM 23127 T for strains 160 T and 143 [18].…”
