On the Mechanism of Constitutive Pdr1 Activator-mediated PDR5 Transcription in Saccharomyces cerevisiae

Abstract: Drug resistance as a result of overexpression of drug transporter genes presents a major obstacle in the treatment of cancers and infections. The molecular mechanisms underlying transcriptional up-regulation of drug transporter genes remains elusive. Employing Saccharomyces cerevisiae as a model, we analyzed here transcriptional regulation of the drug transporter gene PDR5 in a drug-resistant pdr1-3 strain. This mutant bears a gain-of-function mutation in PDR1, which encodes a transcriptional activator for PDR… Show more

“…Pdr5 resembles (orthologous) mammalian MDR1 which is a major factor in tumor resistance [ 29 ]. Regulation of PDR5 is controlled by transcription factors PDR1 and PDR3 and deletion of these transcription factors leads to loss of PDR5 expression, while gain of function mutations in pdr1/3 leads to over expression of this protein [ 30 – 32 ].…”

“…A G648D exchange in UPC2 created an allele that constitutively up-regulated Erg11 expression and thereby improved azole resistance. Similarly, Tac1 in C. albicans and Pdr1 in S. cerevisiae regulate the PDR-type ABC transporter genes Cdr1, Cdr2 , and Pdr5 , respectively [36,71]. In these two yeasts transcription factors mediate the response to azoles by regulating the expression not only of genes for ergosterol biosynthesis but also of efflux transporters.…”

“…Xenobiotic-induced expression of PDR efflux pump genes is complex, but is primarily programmed by the zinc-cluster transcription factor Pdr1 [15][17], along with the orthologous Pdr3 factor [18], [19], which are constitutively bound to PDRE sequences in PDR promoters, even in the absence of toxin [20]. Gene regulatory cofactors including the histone modifier SAGA and chromatin remodeler Swi/Snf have been shown to be recruited to PDR target gene promoters, and implicated in their activation [20].…”

Screens for mutations causing greater PDR1 -dependent drug sensitivity have also indicated the HDAC Rpd3L may have a role in PDR gene activation [21].…”
