Ontogeny of morphine withdrawal in the rat.

Abstract: The present studies examined behavioral changes during precipitated morphine withdrawal in 7- to 42-day-old rat pups. One group of rats was injected with morphine sulfate (10.0 mg/kg) twice daily for 6.5 days. Another group of 7-day-old rats received a lower dose of morphine (3.0 mg/kg). Controls were saline injected or untreated litters (7-day-old pups only). On Day 7, a target pup was injected with saline or naltrexone (0.3-10.0 mg/kg). Preweaning pups were observed in a warm chamber with the litter. Forty-t… Show more

“…Interestingly, the acute morphine withdrawal behavioral repertoire of the 7-day-old rat characterized in the present study is qualitatively comparable of that documented in reports using the chronic morphine injection paradigm (Jones and Barr 1995;Windh et al 1995;. Withdrawal behaviors representative of the acute morphine injection paradigm generally coincide with withdrawal behaviors typical of the chronic morphine administration model.…”

“…The increased withdrawal behaviors included head moves, moving paws, rolling, vocalizations, walking and wall climbing. The categories and intensity of these acute withdrawal behaviors are generally comparable to those seen in withdrawal after chronic morphine administration (Jones and Barr 1995).…”

“…The number of occurrences of each behavior was summed across the 6 minute observation period. Age specific withdrawal behaviors which included head movements, paw movements, rolling, wall climbing, walking, and quiet were scored (Jones and Barr, 1995, Windh et al, 1995, Ceger and Kuhn, 2000. A quiet score was given when the animal was not moving.…”

“…Behaviors during the last 6 out of 8 minutes of video were analyzed by an experimenter blinded to animal treatment. During the assessment period, behaviors were recorded at 15 second intervals on a checklist of age-specific behaviors associated with naloxone precipitated withdrawal following acute or chronic opioid administration in neonatal rats (Jones and Barr, 1995, Windh et al, 1995, Ceger and Kuhn, 2000. The number of occurrences of each behavior was summed across the 6 minute observation period.…”
