Optimization of a direct analysis in real time/time-of-flight mass spectrometry method for rapid serum metabolomic fingerprinting

Abstract: Metabolomic fingerprinting of bodily fluids can reveal the underlying causes of metabolic disorders associated with many diseases, and has thus been recognized as a potential tool for disease diagnosis and prognosis following therapy. Here we report a rapid approach in which direct analysis in real time (DART) coupled with time-of-flight (TOF) mass spectrometry (MS) and hybrid quadrupole TOF (Q-TOF) MS is used as a means for metabolomic fingerprinting of human serum. In this approach, serum samples are first t… Show more

“…A in-depth characterization of the analytical figures of merit of the DART MS approach used here has been recently reported (9), and therefore, the method is only briefly presented. Serum mass spectrometric analysis was done using a DART ion source (IonSense, Inc.) coupled to a JEOL AccuTOF orthogonal time-of-flight (TOF) MS (JEOL, Inc., Japan).…”

“…Liquid chromatography mass spectrometry (LC/MS) is a sensitive technique that can detect many metabolites in a metabolome sample [1]. However, metabolite identification from the mass spectral data alone is often difficult [2][3][4][5][6]. There are only a limited number of metabolite standards available for spectral comparison to identify unknowns.…”

“…For example, the report in 2010 by Zhou et al shows that by applying mono-, di-and tri-methylsilane derivatization, compounds in serum become more volatile and easier to react with precursor ions generated from the DART electrode. 39) As a result, there is a 15-fold increase in signal intensity using this approach. Meanwhile, only analytes reacting with the derivatization reagent have increased intensities.…”
