Optimization of Surfactin Production by Bacillus subtilis Isolate BS5

Abstract: Bacillus subtilis BS5 is a soil isolate that produces promising yield of surfactin biosurfactant in mineral salts medium (MSM). It was found that cellular growth and surfactin production in MSM were greatly affected by the environmental fermentation conditions and the medium components (carbon and nitrogen sources and minerals). Optimum environmental conditions for high surfactin production on the shake flask level were found to be a slightly acidic initial pH (6.5-6.8), an incubation temperature of 30 degrees… Show more

“…Molasses-mineral salts medium (MMSM) is a medium optimized for surfactin production by B. subtilis isolate BS5 that was previously developed by [10]. the composition of MMSM was as follows (per liter): molasses (160 ml); NaNO 3 (5 g), ZnSO 4 (0.16 g), FeCl 3 6H 2 O (0.27 g), MnSO 4 H 2 O (0.017 g), MgSO 4 7H 2 O (0.4 g), NaCl (1 g), KCl (1 g), H 3 PO 4 85% (10 ml), and distilled H 2 O to 1,000 ml, and the pH was adjusted to 7.2 with KOH pellets.…”

“…Molasses of different kinds viz., sugarcane molasses, blackstrap, and beet-molasses are widely used in the fermentation industry for the production of pullulan [22], succinic acid [23], and surfactin [24]. However, the production of SL using this raw material, which is considered much cheaper than others, has not been reported so far.…”

“…1 -Effect of agitation in rhamnolipid production as substrate 19 remained unaffected by aeration rate. On the other hand, Abdel-Mawgoud et al 20 reported that maximum surfactin production by Bacillus subtilis BS5 occurred at an aeration percentage of 90 % and a sharp decline in the production upon decrease of aeration.…”
