Orchestrating stem cell fate: Novel tools for regenerative medicine

Abstract: Mesenchymal stem cells are undifferentiated cells able to acquire different phenotypes under specific stimuli. In vitro manipulation of these cells is focused on understanding stem cell behavior, proliferation and pluripotency. Latest advances in the field of stem cells concern epigenetics and its role in maintaining self-renewal and differentiation capabilities. Chemical and physical stimuli can modulate cell commitment, acting on gene expression of Oct-4, Sox-2 and Nanog, the main stem… Show more

“…The dynamicity generated by medium flow represents the fourth dimension of our system, combining 3D organization and cellular communication. Skin regeneration is a complex process in which skin fibroblasts and stem cells play a crucial role, replacing damaged cellular elements and producing extracellular matrix and hyaluronic acid secretion [39]. Within this context, plants, herb extracts and other bioactive molecules are actually applied as active drugs for tissue regeneration.…”

“…p16 and p19 genes are usually induced when cells are damaged, or after prolonged division, to block cell cycle [26,36]. In particular, p16 inhibits the transition from G1 to S-phase in cell cycle progression [39], exhibiting a fundamental role in homeostasis and aging, maintaining undifferentiated keratinocytes in the deep layers of the tissue. In the skin basal layer, epigenetic modifications repress p16 expression, protecting stem cells from senescence and preserving the regeneration capability of the tissue [39].…”

“…Adipose-derived stem cells (ADSCs) are one of the most promising cells for regenerative medicine, due to their higher differentiation potential and easy propagation, as compared to other sources [ 1 , 2 ]. These cells, located in the stromal vascular fraction (SVF) of adipose tissue, exert a central role during adipogenesis, differentiating to produce mature adipocytes [ 3 ].…”

“…ROS have been recognized as toxic and harmful by-products for stem cells, resulting in DNA damage, premature senescence or cell death [22]. MSCs are largely known for their ability to restore tissue function and differentiate into several phenotypes under appropriate stimuli [23], including osteogenic, chondrogenic, cardiogenic and adipogenic lineages [24][25][26][27]. Nevertheless, as previously demonstrated by Liao N et al, hADSCs expanded for different passages, showing high mRNA levels of p16, p21 and p53 and an increased ROS production, associated with the appearance of a senescent phenotype.…”
