Overview of new developments in and the future of cryopreservation in the laboratory mouse

Abstract: The large-scale mutagenesis programmes underway around the world are generating thousands of novel GA mouse strains that need to be securely archived. In parallel with advances in mutagenesis, the procedures used to cryopreserve mouse stocks are being continually refined in order to keep pace with demand. Moreover, the construction of extensive research infrastructures for systematic phenotyping is fuelling demand for these novel strains of mice and new approaches to the distribution of frozen and unfrozen emb… Show more

“…A great number of genetically engineered mice have been archived in mouse resource banks and are available from the banks via the website of International Mouse Strain Resource [ 1 ]. In the mouse resource bank, various reproductive techniques are used to efficiently preserve, transport, or produce genetically engineered mice [ 2 ]. Until now, a robust and efficient system for managing the mouse resource bank has been established using cryopreservation techniques for sperm, oocytes, and embryos [ 3 6 ].…”

“…At least 54,600 mouse strains have been archived as cryopreserved embryos and sperm [35, 36], which are efficiently distributed to research institutes worldwide. Mice can be rapidly produced and analyzed and then re-archived [37, 38]. Oocyte cryopreservation facilitates the use of archived female germ cells in diverse applications such as introducing gene modifications using IVF with genetically modified sperm or for genome editing after IVF.…”

“…The cryopreservation of preimplantation embryos collected from GM animals allows the long-term storage of valuable lines for an infinite period of time and the subsequent ability to regenerate a colony of live animals in the future (Guan et al, 2012). Cryopreserved embryos are protected from genetic mutation and drift and genetic contamination as well as most facility catastrophes and husbandry expenses are avoided.…”
