Ozone-Induced Programmed Cell Death in the Arabidopsis radical-induced cell death1 Mutant

Abstract: Short, high-concentration peaks of the atmospheric pollutant ozone (O 3 ) cause the formation of cell death lesions on the leaves of sensitive plants. Numerous similarities between the plant responses to O 3 and pathogens suggest that O 3 triggers hypersensitive response-like programmed cell death (PCD). We examined O 3 and superoxide-induced cell death in the O 3 -sensitive radical-induced cell death1 (rcd1) mutant. Dying cells in O 3 -exposed rcd1 exhibited several of the typical morphological characteristic… Show more

“…It has previously been reported that rcd1 is involved in response to a number of abiotic stresses (Overmyer et al, 2000(Overmyer et al, , 2005Ahlfors et al, 2004;Fujibe et al, 2004Fujibe et al, , 2006Katiyar-Agarwal et al, 2006). The rcd1-3 allele behaves similarly to the previously described alleles by displaying increased resistance to chloroplastic reactive oxygen species (ROS) induced by paraquat (Table I).…”

“…rcd1 mutants display accumulation of both ethylene and salicylic acid and altered expression of ethyleneand abscisic acid-responsive genes (Ahlfors et al, 2004). In addition, rcd1 plants are hypersensitive to ozone (Overmyer et al, 2005) and conversely are resistant to UV-B light (Fujibe et al, 2004). The phenotypic analysis suggests that RCD1 may be a convergence point for several hormone signaling pathways involved in the stress response and act both negatively and positively in stress responses (Ahlfors et al, 2004).…”

“…Furthermore, ozone-exposed rcd1 have increased salicylic acid levels and show earlier activation of MPK6 compared to wild-type plants (Overmyer et al, 2005).…”

“…This ethylene response factor was down-regulated (2 fold) in both P. deltoides and the tolerant extreme genotypes but was not differentially expressed in P. trichocarpa and was up-regulated in the sensitive genotypes. The direction of differential expression between both the grandparents and the sensitivity genotypes is particularly interesting considering the role of ethylene in ozone response shown in Overmyer et al (2005) and Tamaoki et al (2003a,b). Low-level ethylene production triggered stress responses more similar to pathogen induced responses while higher levels of ethylene production trigger lesion formation and propagation, a finding that is consistent with the expression changes observed in this study.…”

“…S-adenosyl methionine (SAM) is the precursor molecule to both polyamines and ethylene (Langebartels et al, 1991;Pandey et al, 2000), both of which are thought to be involved in the response to ozone (Langebartels et al, 1991;Overmyer et al, 2005). It is of interest, therefore, that SAM Synthetase (SAMS) co-located to QTL for both late and early season visible damage on LG II.…”
