PACSINs Bind to the TRPV4 Cation Channel

Abstract: TRPV4 is a cation channel that responds to a variety of stimuli including mechanical forces, temperature, and ligand binding. We set out to identify TRPV4-interacting proteins by performing yeast two-hybrid screens, and we isolated with the avian TRPV4 amino terminus the chicken orthologues of mammalian PACSINs 1 and 3. The PACSINs are a protein family consisting of three members that have been implicated in synaptic vesicular membrane trafficking and regulation of dynamin-mediated endocytotic processes. In bi… Show more

“…9A), and the combined mutation of prolines at positions 142 and 143 to alanine and leucine, respectively (mutant isoform P142A/P143L), has previously been shown to abolish the interaction with PACSIN 3 (34). In [Ca 2ϩ ] i imaging experiments, the P142A/ P143L mutant isoform responded to stimulation with 4␣-PDD, heat, HTS, or AA independent of the presence of PACSIN 3 (Figs.…”

“…Heat-induced Activation of TRPV4-In a previous study it was shown that interaction with PACSIN 3 affects the subcellular distribution of TRPV4, which results in an apparent increase of the relative plasma membrane association of the channel (34). It remained untested whether PACSIN 3 affects channel gating.…”

“…More recent studies indicate that TRPV4 is not only activated by osmotic stimuli but also by warm temperatures, the phorbol derivative 4␣-phorbol-12,13-didecanoate (4␣-PDD), and lipid products of the arachidonic acid cascade (10 -13). In addition to these, TRPV4 has been reported to have an association with many proteins (PACSIN3 (14), OS9 (15), microtubule-associated protein 7 (16), inositol 1,4,5-trisphosphate receptor 3 (17), aquaporin 5 (18), TRPP2 (19), caveolin 1 (20), CFTR chloride channel (21), and BK channel (22)). It reminds us that TRPV4 should be flexible to interact with a variety of proteins.…”
