“…In addition, several lines of evidence imply that it is crucial for the formation of MDCK epithelial cysts to regulate epithelial cell polarity via the PAR3/aPKC/PAR6 complex and its downstream CRB3/PALS1/PATJ complex in the apical cortical domains (Hurd et al, 2003;Lemmers et al, 2004;Roh et al, 2003). Our model is further supported by the observation that par3 RNAi in C. elegans disturbs the apical organization of distal spermathecal cells (Aono et al, 2004). Taken together, our present data suggest that PAR3 is responsible for epithelial cyst formations by interpreting polarity cues from the cell-cell and cell-ECM interactions that lead to the establishment of apical cortical domains.…”

“…The rrf-1 mutants are defective with RNAi in the somatic cells but not in the germ cells (Sijen et al, 2001). Thus, if function of an RNAi target is important in the somatic gonad (either sheath contraction [McCarter et al, 1997] or spermathecal dilation [Clandinin et al, 1998;Aono et al, 2004]), ovulation will be defective in wild-type but normal in the rrf-1 background.…”

“…Examining the function of PAR-3 in C. elegans epithelial cells has been hindered by the essential requirement of maternal PAR-3 in polarizing the one-cell embryo. Existing mutations in the single par-3 gene eliminate maternal but not zygotic PAR-3 protein and cause a maternal-effect lethal phenotype (Aono et al, 2004;Etemad-Moghadam et al, 1995;Kemphues et al, 1988); mutant embryos have scrambled cell fates and fail to form organized epithelia. The function of PAR-3 in epithelial cells that form in larvae has been examined by feeding par-3 double-stranded (ds) RNA to freshly hatched worms, reducing levels of zygotically expressed PAR-3.…”
