Partial Validation of Ultra Performance Liquid Chromatography Method for Quantification of Isoniazid-Pyrazinamide in Human Samples

Abstract: Isoniazid (INH) and pyrazinamide (PZA) are first-line drugs in the treatment of meningitis of tuberculosis, in addition to rifampicin. The use of these drugs will determine the success of therapy to kill Mycobacterium tuberculosis in brain tissue that is difficult to penetrate by other drugs. Therefore, it is necessary to conduct research and monitor the use of this drug in plasma and cerebrospinal fluid (CSF) patients with TBM. This study aimed to determine the method of analysis of INH and PZA using Ultra Pe… Show more

“…Bioanalysis of isoniazid was performed at the Pharmacokinetic Laboratory of the Faculty of Medicine of the Universitas Padjadjaran using a validated UPLC method. 20 , 21 PK parameters of isoniazid were assessed non-compartmentally with the PKNCA package (version 0.9.5) in RStudio for macOS. Main PK measures included area under the plasma concentration–time curve from 0 to 24 h after dosing (AUC 0–24 ) and peak concentration ( C max ).…”
