Pathobiological Interactions of Local Bone Marrow Renin-Angiotensin System and Central Nervous System in Systemic Arterial Hypertension

Abstract: Circulating renin-angiotensin system (RAS) and local paracrin-autocrin-intracrin tissue-based RAS participate in numerous pathobiological events. Pro-inflammatory, pro-fibrotic, and pro-thrombotic consequences associated with local RAS activation have been detected at cellular and molecular level. Regenerative progenitor cell therapy in response to RAS modulating pharmacotherapy has emerged as an adjunct in the context of endothelial cell injury and regeneration to improve regeneration of the vascular endothel… Show more

“…The use of dihydropyridine calcium channel blockers and renin–angiotensin system inhibitors (RASi) would be preferable due to the strong selectivity for the vascular compartment ( Hayman et al, 2012 ). In addition, the bone marrow RAS is finely implicated in the development of hypertension ( Ciftciler and Haznedaroglu, 2020 ). In fact, recently, RAS inhibitors (RASi) have been associated with a reduction in CV events in CML patients treated with NGTKI ( Mulas et al, 2020 ).…”

“…At rst, it was thought that RAS was just an endocrine system composed of substrates or enzymes from different tissues, such as renin from kidney, angiotensinogen from liver and ACE from lung [10]. Then relatively independent RAS in the heart, kidney, liver, lung, pituitary, digestive tract, muscle tissue, tumor tissue, immune system and central nervous system have been gradually found, which could play a local role in many organs and tissues through paracrine/autocrine [14]- [20]. Of course, this also includes the reproductive system.…”

“…Locally active RAS in the BM that affects the growth, production, proliferation, and differentiation of hematopoietic cells. The interactions of hematopoietic RAS with other tissue RASs are evident (10,11,22,23). All of the major RAS molecules including renin, angiotensinogen, angiotensin receptors and ACE are located within the BM microenvironment.…”
