PCR arrays indicate that the expression of extracellular matrix and cell adhesion genes in human adipocytes is regulated by IL-1β(interleukin-1β)

Abstract: The role of IL-1β in regulating the expression of extracellular matrix (ECM) and cell adhesion genes in human adipocytes has been examined. Adipocytes differentiated in culture were incubated with IL-1β for 4 or 24 h and RNA probed with PCR arrays for 84 ECM and cell adhesion genes. Treatment with IL-1β resulted in changes in the expression at one or both time points of ∼50% of the genes probed by the arrays, the majority being down-regulated. Genes whose expression was down-regulated by IL-1β included those e… Show more

“…IL-1β, a leukocytic pyrogen activated by macrophages, is involved in the inflammatory process, and its expression in the regulation of ECM and cell adhesion has been examined in human adipocytes. The incubation with IL-1β in adipocytes upregulates ICAM1 and VCAM1, indicating an effect on the expression of ECM and cell adhesion genes in human adipocytes, consistent with the derangement of tissue structure during inflammation in adipose tissue [151]. A whole-genome expression profile of liver, SM, SAT, and VAT was carried out in metabolically healthy obese and metabolically unhealthy individuals by using the weighted gene co-expression network analysis (WGCNA) to build withinand inter-tissue gene networks.…”

“…The expressions of IL-1b and its receptor inhibition were proved to promote ECM repair and prevent IDD [40]. IL-1b has the ability to affect MMP-3, ECM at the gene level and in catabolic mediators [41]. Therefore, inhibiting the action of IL-1b or its receptor antagonist may be effective in preventing or even reversing the intervertebral disc degeneration.…”

“…MMP-3 (stromelysin I) is a proteoglycanase and a key player in ECM degradation and remodeling [ 4 ]. MMP-3 was also associated with the inflammatory response during adipose tissue expansion, as well as with higher fat tissue levels, indicating a role of MMP-3 in adipogenesis [ 27 ]. According to the literature, alterations of MMP-3 pathway in obesity could be related to functional SNPs in MMP-3 gene.…”
