Periostin facilitates eosinophil tissue infiltration in allergic lung and esophageal responses

Abstract: Periostin is an extracellular matrix protein that has been primarily studied in the context of the heart, where it has been shown to promote cardiac repair and remodeling. In this study, we focused on the role of periostin in an allergic eosinophilic inflammatory disease (eosinophilic esophagitis (EE)) known to involve extensive tissue remodeling. Periostin was indeed markedly overexpressed (35-fold) in the esophagus of EE patients, particularly in the papillae, compared with control individuals. Periostin exp… Show more

“…Accordingly, recombinant periostin was found to directly enhance adhesion of eosinophils and monocytes to fibronectin and function as a chemoattractant to primary monocytes and macrophages in migration assays. 24,25 Chronic inflammation plays a central role in development of fibrosis and kidney damage. Concomitantly to the attenuation of inflammation, periostin deficiency diminished the expression of collagens and TGF-b1, and improved renal structure and function.…”

“…Although periostin has been considered a mediator of inflammation and fibrosis in allergic diseases, these studies have focused on periostin derived from fibroblasts (22,23); to our knowledge, the expression of periostin by epithelial cells has not been considered in mechanisms of airway fibrosis in asthma.…”

“…These inflammatory mediators are known to induce various effects such as up‐regulation of adhesion molecules to modulate cellular trafficking and recruitment, mucus secretion, tissue remodeling, and impairment of barrier integrity,24, 25 which can contribute to disease progression. For example, eosinophil‐derived TGF‐β is linked to tissue remodeling and induction of the extracellular matrix protein periostin in patient biopsy specimens which increases eosinophil infiltration in the mucosal layer, thus facilitating disease pathogenesis 26, 27. Furthermore, as indicated above, granule proteins such as eosinophil cationic protein, Epx, and major basic protein released by activated eosinophils during inflammation can contribute to tissue damage (alter barrier function) and dysfunction (diarrhea with bleeding) 9, 10, 11, 12.…”
