Peripheral CD4 T-Cell Depletion Is Not Sufficient to Prevent Ischemic Acute Renal Failure

Abstract: Ischemic ARF can occur in the absence of classical T-cell function. The evaluation of other inflammatory mediators (e.g., macrophages or NK cells) as a source of IL-18 and mediator of ischemic ARF warrants further investigation.

“…TCRαβ deficient mice had significantly improved renal function with reduced TNF-α and IL-6 production after IRI, while TCRγδ deficient mice only presented slight amelioration [60]. While initially another group failed to observe a role of TCR in renal IRI [61], the importance of TCR in kidney IRI was confirmed by careful knockout and antibody depletion studies [62].…”

“…Rag1 -/-mice lack T and B cells and are not protected from AKI induced by ischemia (55,58). Multiple groups have detected no difference in serum creatinine 24 hours after IRI in mice with depletion of either TCRαβ or TCRγδ (55,59,60), although some have found reduced structural damage (60,61) and/or improved survival and lower serum creatinine (60) at later time points after reperfusion.…”

“…For all procedures, mice were anesthetized with IP Avertin (2,2,2-tribromoethanol; Aldrich, Milwaukee, WI), a midline incision was made, and the renal pedicles were identified. In the ischemic ARF group, both renal pedicles were clamped for 22 min (23,24). In the unilateral ischemia model, the left renal pedicle was clamped for 22 min.…”
