Phage-displayed antibody fragments recognizing dengue 3 and dengue 4 viruses as tools for viral serotyping in sera from infected individuals

Abstract: The current study shows the usefulness of dengue-3- and dengue-4-specific phage-displayed antibody fragments as tools for viral detection and serotyping in sera from infected individuals. C6/36 HT cells were inoculated with acute-phase sera from patients, and supernatants were collected daily and analyzed by ELISA using phage-displayed antibody fragments as serotype-specific detector reagents. Serotyping of most samples was possible as early as two to three days postinoculation. Results were comparable with th… Show more

“…Phage antibody libraries have been widely used to select antibodies against infectious agents. These include antibodies that discriminate between different strains of Hantavirus71, Dengue virus72, influenza73,74, Ebola75 and Venezuelan equine encephalitis virus76. Given that many of these viruses are classified serologically, the ability to select phage antibodies with similar specificities is not surprising, but unlike antibodies generated by immunization, these have the potential to be used therapeutically.…”

“…Using an anti-PVIII antibody to detect bound phages interacting with an immobilized partner is thus a very effective amplification system to detect weak interactions. Such potential has been exploited by using whole phages as detector particles in diagnosis (Cabezas et al, 2009). The current work, aimed at detecting the moderate affinity interaction of IL-2 with the isolated alpha chain, as well as the even weaker residual binding of the mutated IL-2 variants, took advantage of this extremely high sensitivity to circumvent our previous failure to measure alpha chain binding to IL-2 variants (unpublished results).…”

“…This adaptable tool fuels innovative strategies for disease detection and the exploration of biomarkers. Once engineered, these modified phages are useful in diverse diagnostic applications, offering sensitive and precise detection of disease-related entities in clinical samples, ranging from minute targets such as viruses to substantial entities such as cancer cells ( Table 1 ) [ 182 , 183 , 184 , 185 , 186 ].…”
