Phage vB_BsuP-Goe1: the smallest identified lytic phage ofBacillus subtilis

Abstract: Currently, interest in phage science is on the rise again as this subject remains largely unexplored, and its potential diversity mainly untapped. Here, we present phage vB_BsuP-Goe1, a new isolate that infects Bacillus subtilis The phage forms round plaques with a matt outline on agar plates containing a B. subtilis Δ6 lawn. Transmission electron microscopy and genomic analyses revealed that phage vB_BsuP-Goe1 belongs to the Podoviridae family as a new member of the Phi29likevirus genus. The phage genome comp… Show more

“…Phages of the φ29-like group are the smallest Bacillus infecting phages (for review see [ 7 ]) and members of the Podoviridae family. The sequenced representatives harbour a small linear genome (18.4–21.1 kbp [ 8 ]) covalently linked to a terminal protein (TP) at the 5′ genome end. A DNA replication mechanism first investigated in this phage group has attracted particular attention: Protein-primed genome replication, is initiated at the origin at the 3′ end of the genome together with a TP protein by forming a TP-dAMP complex, using the second base of the template.…”

“…To verify whether the pJOE8999 vector system [ 23 ] is suitable for in vivo genetic engineering of virulent phages, a proof of concept experiment with the smallest known B. subtilis phage Goe1 [ 24 ] was performed. The gene Goe1_c00180 coding for the head fiber protein was selected as target.…”

“…The virulent phage vB_BsuP-Goe1 (Goe1) [ 24 ] serves as model phage for this investigation. It reveals Podoviridae morphology and is a member of the genus Phi29virus [ 24 ]. It has a genome of 18,379 bp and is closely related to the well-understood phage phi29 (63.7% global nucleotide similarity).…”

“…Raw sewage from a municipal sewage treatment plant in Göttingen (Germany, 51°33′15.4″ N 9°55′06.4″ E) was used as inoculum. The isolation and propagation procedure were performed as described previously [18]. Plaque assays were performed by mixing 100 µL of the overnight host culture with 100 µL of virus suspension and incubating for 5 min at room temperature.…”
