Pharmacological Ascorbate as an Adjuvant for Enhancing Radiation-Chemotherapy Responses in Gastric Adenocarcinoma

Abstract: Gastric adenocarcinoma most often presents at an advanced stage and overall five-year survival of ∼30%. Pharmacological ascorbate (high-dose IV ascorbate) has been proposed as a promising nontoxic adjuvant to standard radio-chemotherapies in several cancer types. In the current study, pharmacological ascorbate (0.5-2 m M) caused a dose-dependent decrease (70-85% at 2 m M) in clonogenic survival of gastric adenocarcinoma cells (AGS and MNK-45), but was relatively nontoxic to a small intestinal epithelial nonimm… Show more

“…These same genes have also been nominated as candidate genes for alcohol dependence in a human genetic association study (Zuo et al, 2011 ). We further confirmed that Ptp4a1 is a candidate gene in stress and alcohol responses (Baker et al, 2017 ) by using differentially expressed (DE) genes and QTL analysis in BXD recombinant inbred (RI) strains. In addition, we identified a sequence variant in a circadian rhythm gene, Per3 , and associated it with stress and alcohol responses by integrating expression QTL (eQTL) and phenotype QTL analyses (Wang et al, 2012 ).…”

“…In addition, G-protein alpha ( Gna1 , Gnal2 , Gnb1 and Gng2 ) and its regulators ( Rgs10 and Rgs14 ) are included in this subnetwork. The involvement of these genes in both alcohol and stress responses is well known (Baker et al, 2017 ).…”

“…In reality, against the advances in the classic tumor chemotherapy, the prediction chance of the advanced gastric cancer in patients is yet very low. Agent 5-FU, being anti-neoplastic in nature, is the first-line chemotherapy having a wide range of activity in gastro-intestinal tumors; however, like the majority of tumor medicines, its major disadvantage is the high level of toxicity and the low rate of therapeutic effect (O'Leary et al, 2018;Shah and Schwartz, 2001). As a result, it is urgently required that new therapeutic modalities be introduced for the gastric tumor.…”

“…Experiments performed in vitro to test compatibility with other anti-carcinogenic substances revealed that AA can have a synergistic effect with some of them (Espey et al, 2011;Ma et al, 2014;Hatem et al, 2018;O'Leary et al, 2018;Graczyk-Jarzynka et al, 2019). For instance, Gemcitabine in combination with AA (Espey et al, 2011) have a synergistic cytotoxic effect in eight pancreatic cancer cell lines, which is mediated by the pro-oxidant effect of ascorbate, again with an increase in the production of H 2 O 2 .…”
