Pharmacological Interventions for the MATRICS Cognitive Domains in Schizophrenia: What’s the Evidence?

Abstract: Schizophrenia is a disabling, chronic psychiatric disorder with a prevalence rate of 0.5–1% in the general population. Symptoms include positive (e.g., delusions, hallucinations), negative (e.g., blunted affect, social withdrawal), as well as cognitive symptoms (e.g., memory and attention problems). Although 75–85% of patients with schizophrenia report cognitive impairments, the underlying neuropharmacological mechanisms are not well understood and currently no effective treatment is available for these impair… Show more

“…A metaanalysis revealed that medication targeting the cholinergic system result in marginal improvement in verbal and spatial learning and memory (154). Preliminary evidence also suggest that oxytocin may improve social cognition and verbal learning and memory (155), but the efficacy of such intervention has proven to be inconclusive and more research is still needed (101).…”

“…In contrast, negative symptoms include social withdrawal and anhedonia, while cognitive symptoms involve deficits in attention, working memory, and impulse control (Lewis et al 2005). To date, no available treatment or combination of treatments reduces symptom presentation across all three domains (Vingerhoets et al 2013). Importantly, cognitive symptoms are highly indicative of patient functional outcome, including community functioning and interpersonal interactions, implicating them in patient quality of life (Bhagyavathi et al 2015).…”

“…It is becoming increasingly clear that the development of treatments for the cognitive symptoms of schizophrenia requires urgent attention (Minzenberg and Carter 2012;Schaefer et al 2013;Vingerhoets et al 2013). To achieve this aim we need valid animal models of the cognitive impairments in schizophrenia (Nestler and Hyman 2010).…”
