Phospholipase A2 Biochemistry

Abstract: The phospholipase A 2 (PLA 2 ) superfamily consists of many different groups of enzymes that catalyze the hydrolysis of the sn-2 ester bond in a variety of different phospholipids. The products of this reaction, a free fatty acid, and lysophospholipid have many different important physiological roles. There are five main types of PLA 2 : the secreted sPLA 2 's, the cytosolic cPLA 2 's, the Ca 2+ independent iPLA 2 's, the PAF acetylhydrolases, and the lysosomal PLA 2 's. This review focuses on the superfamily … Show more

“…cPLA 2 ␣ can be activated by several mechanisms (reviewed in Refs. 11,12). It contains a Ca 2ϩ binding domain (C2) that allows interaction with membrane phospholipids in response to increased intracellular Ca 2ϩ concentrations, and the single substitution D43N in this domain abrogates the Ca 2ϩ -dependent translocation of cPLA 2 ␣ to membranes (13)(14)(15).…”

“…In this regard, anionic phospholipids like phosphatidylinositol 4,5-bisphosphate (PtdIns-4,5-P 2 ) (20, 21) and ceramide 1-phosphate (Cer-1-P) (22)(23)(24)(25) bind specific regions of the enzyme and decrease its Ca 2ϩ requirement to interact with membranes. Also, phosphorylation of cPLA 2 ␣ at Ser residues 505, 515, and 727 appears to play a role in enzyme activation (11,12). Among these sites, it is generally accepted that Ser-505 is the one that leads to increased catalytic activity (26), and phosphorylation of Ser-505 depends on MAPK, among which most reports implicate ERK and p38 (11,12).…”

“…Phospholipases A2 (PLA2s) constitute a superfamily of enzymes that catalyze the hydrolysis of fatty acids from the sn-2 position of glycerophospholipids to produce free fatty acids and lysophospholipids, which are both involved in signaling transduction and metabolic processes, and account for a vast number of diseases [1,2] . PLA2s, then, are obvious candidates for pharmacological research and intervention.…”
