Physicochemical, Pharmacokinetic, and Toxicity Evaluation of Soluplus® Polymeric Micelles Encapsulating Fenbendazole

Abstract: Fenbendazole (FEN), a broad-spectrum benzimidazole anthelmintic, suppresses cancer cell growth through various mechanisms but has low solubility and achieves low blood concentrations, which leads to low bioavailability. Solubilizing agents are required to prepare poorly soluble drugs for injections; however, these are toxic. To overcome this problem, we designed and fabricated low-toxicity Soluplus® polymeric micelles encapsulating FEN and conducted toxicity assays in vitro and in vivo. FEN-loaded Soluplus® mi… Show more

“…21 Despite its numerous anticancer effects, clinical application of FEN is difficult owing to its low solubility in water. 22,23 Rapamycin (RAPA) is a carboxylic lactone-lactam macrolide antibiotic that was originally isolated from the bacterium Streptomyces hygroscopicus. 24,25 It was initially developed as an antifungal agent.…”

“…However, MUC-30 was somewhat more cytotoxic than MUC-30-loaded micelles, probably because MUC-30 can be transported into the nucleus of cells by the passive diffusion mechanism, while the drug-loaded micelles have to be internalized by endocytosis, release the loaded drugs, and then, diffuse through the endocytic before entering to nucleus of cells [ 32 ]. However, the properties of the drug-loaded nanoparticles were found to be safer with normal cells (L929) than those used with free drugs [ 33 ], which results in normal cell death as well.…”

“…In the present study, we propose to use 2-(2′-pyridyl)benzimidazole (Hpben) as a ligand with potentially three N-donor atoms (see Scheme 1 B), which would lead to different coordinations with the gold atom [ 35 ]. Benzimidazoles are very versatile ligands that show various pharmacological properties such as antibacterial, antiviral, and anti-inflammatory [ 37 , 38 ]. Our aim is to obtain N-Au-P complexes, for which Hpben and two different phosphines are used: triphenylphosphine and triethylphosphine.…”
