In the present study, different types of gibberellins (GAs) in the culture filtrate (CF) of Photorhabdus temperata M1021 were quantified. The analysis of CF helped in profiling various bioactive GAs: GA 1 , GA 3 , GA 4 , and GA 7 . Several physiologically inactive GAs: GA 9 , GA 12 , and GA 20 were detected as well. Siderophore production was also investigated by growing P. temperata M1021 on chrome azurol-S blue agar plates. Furthermore, the strain was inoculated into 'Waito-C' (Oryza sativa L.) rice plants, which significantly (P < 0.05) increased plant growth attributes such as plant length, chlorophyll content, and fresh and dry biomass compared with those in controls. In a separate experiment, canola (Brassica napus L.) seeds treated with CF of M1021 were significantly (P < 0.05) accelerated germination rate as well as biomass production. Findings of the present study suggest that the strain M1021 contributes an important role in the plant growth by synthesizing a wide array of bioactive metabolites.

Photorhabdus temperata M1021 is an entomopathogenic enterobacterium, producing an array of metabolites in the culture extract. Photorhabdus spp. produced various classes of toxic proteins and secondary metabolites, which generate mortality in a wide range of insects. Aim of present study was to assess the effects these metabolites towards the insect and plants. Intra-hemocoel injections of whole cell and soluble extract caused 100% mortality in Galleria mellonella larvae within 48 h as compare to control. However, the culture filtrate (CF) took 72 h to kill 100 % larvae. In addition, there was no significant (P<0.05) decrease in the rate of larval mortality, injected with CF, treated at 70°C for 30 min as well as with 50 mg/l proteinase-K. Gas chromatography mass spectrometry analysis revealed 23 compounds, among which "trans-Cinnamic acid" and "indole" were the most important for the insecticidal activities and plant growth promotion. The auxin contents quantified through Salkowski's assay were 28 μg/ml to 30 μg/ml, insignificantly varied the by the addition of L-tryptophan ranged from 0 g/l to 0.5 g/l. The optimum pH for the auxin production was determined to be pH 7.0. The CF treatment increases significantly (P<0.05) plant growth that attributes e.g. total plant length, chlorophyll content and biomass of both Dongjin-beyo and Waito-Crice plants, in a comparison with control. The Photorhabdus temperata M1021 secretes toxic compounds that kill their insect hosts by arresting immune responses. Present study was aimed to purify the insecticidal and antimicrobial compound(s) from the culture extract of P. temperata M1021 through bioassay guided fractionation. An ethyl acetate (EtOAc) extract of the P. temperata M1021 exhibited 100% mortality in Galleria mellonella larvae within 72 h. In addition, EtOAc extract and bioactive compound 1 purified form the extract through to column chromatography, showed phenol oxidase inhibition up to 60% and 80% respectively. The analysis of (1)H and (13)C NMR spectra revealed the identity of pure compound as "benzaldehyde". The benzaldehyde showed insecticidal activity against G. mellonella in a dose-dependent manner and 100% insect mortality was observed at 108 h after injection of 8 mM benzaldehyde. In a PO inhibition assay, 4, 6, and 8 mM concentrations of benzaldehyde were found to inhibit PO activity about 15%, 42%, and 80% respectively. In addition, nodule formation was significantly (P < 0.05) inhibited by 4, 6, and 8 mM of benzaldehyde as compare to control. Moreover, benzaldehyde was found to have great antioxidant activity and maximum antioxidant activity was 52.9% at 8 mM benzaldehyde as compare to control. Antimicrobial activity was assessed by MIC values ranged from 6 mM 10 mM for bacterial strains and 8 mM to 10 mM for fungal strains. The results suggest that benzaldehyde could be applicable for developing novel insecticide for agriculture use.
