Phytochromes A and B Mediate Light Stabilization of BIN2 to Regulate Brassinosteroid Signaling and Photomorphogenesis in Arabidopsis

Abstract: Phytochromes A and B (phyA and phyB) are the far-red and red lights photoreceptors mediating many light responses in Arabidopsis thaliana. Brassinosteroid (BR) is a pivotal phytohormone regulating a variety of plant developmental processes including photomorphogenesis. It is known that phyB interacts with BES1 to inhibit its DNA-binding activity and repress BR signaling. Here, we show that far-red and red lights modulate BR signaling through phyA and phyB regulation of the stability of BIN2, a glycogen synthas… Show more

“…The BR pathway intersects with light and ABA signalling at many levels. Light is known to inhibit BR biosynthesis, to stabilize or activate BIN2 and to impact BR-responsive transcriptional regulation [53][54][55][56][57]; reviewed by [36,58,59]. BIN2 modulates light responses by phosphorylating PIFs, transcriptional regulators inhibited by phytochromes, thereby targeting them for degradation [60,61].…”

“…PhyA controlling the stability of light-responsive genes is well studied. However, how phyA rapidly controls the transcription of light-responsive transcription factors remains largely unknown ( 46 , 47 ). It is necessary to explore whether other phyA-activated light-responsive genes exhibit similar characteristics and whether this is a universal mechanism for the pattern of gene expression regulated by phyA signaling after etiolated seedlings are exposed to light.…”

“…Notably, in the plant hormone transduction pathway of the iSAS-root based on RNA-seq analysis, three genes were obviously upregulated. Among them, BIN2 has been proven to directly interact with phyB and regulate BR signaling and photomorphogenesis in Arabidopsis ( Zhao et al, 2022 ). The hypocotyls of BIN2 bin2-1 gain-of-function mutants were found to be shorter than those of the wild type under R and FR light.…”
