Improvement of the bioavailability of poorly soluble medicinal substances is currently one of the major challenges for pharmaceutical industry. Enhancing the dissolution rate of those drugs using novel methods allows to increase their bioavailability. In recent years, silica-based mesoporous materials have been proposed as drug delivery systems that augment the dissolution rate. The aim of this study was to analyse the influence of phenylbutazone adsorption on SBA-15 on its dissolution rate. Moreover, we examined the cytotoxicity of the analyzed silica. The material was characterized by SEM, TEM, DSC, 1H-NMR, XRD, and FT-IR. The phenylbutazone did not adsorb on unmodified SBA-15, while the adsorption on APTES-modified SBA-15 resulted in 50.43 mg/g of loaded phenylbutazone. Phenylbutazone adsorbed on the APTES-modified SBA-15 was then released in the hydrochloric acidic medium (pH 1.2) and phosphate buffer (pH 7.4) and compared to the dissolution rate of the crystalline phenylbutazone. The release profiles of the amorphous form of adsorbed phenylbutazone are constant in different pH, while the dissolution rate of the crystalline phenylbutazone depends on the pH. The cytotoxicity assays were performed using the Caco-2 cell line. Our results indicate that the analyzed material ensured phenylbutazone adsorption in an amorphous state inside the mesopores and increased its dissolution rate in various pH levels. Furthermore, the cytotoxicity assay proved safety of studied material. Our study demonstrated that APTES-modified SBA-15 can serve as a non-toxic drug carrier that improves the bioavailability of phenylbutazone.

Both cyclodextrin (CD) and porous silica possess interesting properties of adsorption and release. A silica–CD hybrid, therefore, could synergically merge the properties of the two components, giving rise to a material with appealing properties for both environmental and pharmaceutical applications. With this aim, in the present study, a first hybrid is obtained through one-pot sol–gel synthesis starting from CD and tetramethyl orthosilicate (TMOS) as a silica precursor. In particular, methyl-β-cyclodextrin (bMCD) is selected for this purpose. The obtained bMCD–silica hybrid is a dense material containing a considerable amount of bMCD (45 wt.%) in amorphous form and therefore represents a promising support. However, since a high specific surface area is desirable to increase the release/adsorption properties, an attempt is made to produce the hybrid material in the form of an aerogel. Both the synthesis of the gel and its drying in supercritical CO2 are optimized in order to reach this goal. All the obtained samples are characterized in terms of their physico-chemical properties (infra-red spectroscopy, thermogravimetry) and structure (X-ray diffraction, electron microscopy) in order to investigate their composition and the interaction between the organic component (bMCD) and the inorganic one (silica).

Background: High-performance thin-layer chromatography (HPTLC) is an advantageous, modern analytical technique based on the principles of thin-layer chromatography (TLC), which provides essential benefits, such as improved sample application, better and faster separation, and less mobile phase usage. The aim of this work was to develop a simple and rapid HPTLC method that could be used for the identification of meloxicam and piroxicam. Methods: HPTLC. An analysis was carried out using silica gel 60 F254 glass TLC plates and as the mobile phase: hexane: ethyl acetate: glacial acetic acid, in a ratio of 65:30:5 v/v/v. For the standard solution preparation, ethanol was used. Front: 60 mm. Detection was performed at 366 nm. Results: The Rf value for meloxicam was 0.81 and the Rf value for piroxicam was 0.57. The proposed method can be used in the detection of the analyzed compounds in very low concentrations. It was established that the detection limit of meloxicam was 0.04 μg per band and that of piroxicam was 0.05 μg per band. It was also established that the quantitation limit of meloxicam was 0.12 μg per band and that of piroxicam was 0.15 μg per band. Conclusions: The proposed method is simple, sensitive, stable, cost effective, and eco-friendly. It could be used in research or for routine quality control purposes.
