Osteoporosis is a frequent complication of chronic inflammatory diseases and increases in the pro-inflammatory cytokines make an important contribution to bone loss by promoting bone resorption and impairing bone formation. Omentin-1 is a newly identified adipocytokine that has anti-inflammatory effects, but little is known about the role of omentin-1 in inflammatory osteoporosis. Here we generated global omentin-1 knockout (omentin-1−/−) mice and demonstrated that depletion of omentin-1 induces inflammatory bone loss-like phenotypes in mice, as defined by abnormally elevated pro-inflammatory cytokines, increased osteoclast formation and bone tissue destruction, as well as impaired osteogenic activities. Using an inflammatory cell model induced by tumor necrosis factor-α (TNF-α), we determined that recombinant omentin-1 reduces the production of pro-inflammatory factors in the TNF-α-activated macrophages, and suppresses their anti-osteoblastic and pro-osteoclastic abilities. In the magnesium silicate-induced inflammatory osteoporosis mouse model, the systemic administration of adenoviral-delivered omentin-1 significantly protects from osteoporotic bone loss and inflammation. Our study suggests that omentin-1 can be used as a promising therapeutic agent for the prevention or treatment of inflammatory bone diseases by downregulating the pro-inflammatory cytokines.

Psoriasis is a chronic skin disease characterized by systemic inflammation associated with an increased risk of metabolic and cardiovascular complications. A special role in maintaining systemic inflammation is attributed to obesity. Adipose tissue is an active endocrine organ which, through adipokine secretion, is involved in the regulation of many systemic metabolic pathways. Obesity increases the risk of developing type 2 diabetes mellitus, atherosclerosis, cardiovascular diseases and hypertension. The paper presents the characteristics of two relatively recently discovered and little known adipokines: omentin and vaspin. They are considered to have beneficial effects preventing the development of metabolic disorders associated with obesity. In addition, attention was given to their possible involvement in the pathogenesis of psoriasis and potential use as markers of disease severity and the risk of developing metabolic complications. stReszczenIe Łuszczyca jest przewlekłą chorobą skóry, w której ogólnoustrojowy stan zapalny wiąże się ze zwiększonym ryzykiem rozwoju powikłań metabolicznych i sercowo-naczyniowych. Szczególne znaczenie w podtrzymaniu stanu zapalnego w organizmie ma otyłość. Tkanka tłuszczo-wa jest aktywnym organem endokrynnym, który poprzez wydzielane adipokiny bierze udział w regulacji licznych szlaków metabolicznych ustroju. Otyłość zwiększa ryzyko rozwoju cukrzycy typu 2, miażdżycy, chorób sercowo-naczyniowych oraz nadciśnienia. W pracy przedstawiono charakterystykę stosunkowo nowo odkrytych i mało poznanych adipokin: omentyny i waspiny, którym przypisuje się korzystne działanie, chroniące przed rozwojem zaburzeń metabolicznych towarzyszących otyłości. Ponadto zwrócono uwagę na ich potencjalny udział w patogenezie łuszczycy oraz możliwość wykorzystania jako markerów nasilenia choroby oraz ryzyka rozwoju powikłań metabolicznych.

The Relationship Between Circulating Neuregulin-1 and Coronary Collateral Circulation in Patients with Coronary Artery Disease

Coronary collateral circulation (CCC) plays a crucial role in myocardial blood supply, especially for ischemic myocardium. Previous study has shown that neuregulin-1 is a prominent angiogenic factor in diabetic cardiomyopathy, whereas the relationship between neuregulin-1 and CCC has not been investigated. Thus, we aimed to investigate relationship between circulating neuregulin-1 levels and CCC in stable coronary artery disease patients. Coronary artery disease patients with a stenosis of !90% as evidenced by coronary angiography were included in our study. According to the Rentrop-Cohen classification, coronary collateral degree was graded as 1 to 4. Patients with collateral degree grade 0 or 1 were enrolled in poor CCC group, whereas patients with grade 2 or 3 were enrolled in good CCC group. Plasma neuregulin-1 level was significantly increased in good collateral group and positively related to Rentrop grade (P < 0.01). Multivariate regression analysis and ROC (receiver operating characteristic curve) revealed that plasma neuregulin-1 could predict CCC status effectively. Increased plasma neuregulin-1 level was related to better CCC in patients with coronary artery disease. Neuregulin-1 was an independent and reliable predictor for good coronary collateral development and provided a potential therapeutic strategy to reduce myocardial ischemia injury.
