Plasma proteome changes in subjects with Type 2 diabetes mellitus with a low or high early insulin response

Abstract: Circulating proteins contribute to the pathogenesis of T2DM (Type 2 diabetes mellitus) in various ways. The aim of the present study was to investigate variations in plasma protein levels in subjects with T2DM and differences in beta-cell function, characterized by the EIR (early insulin response), and to compare these protein levels with those observed in individuals with NGT (normal glucose tolerance). Ten subjects with NGT+high EIR, ten with T2DM+high EIR, and ten with T2DM+low EIR were selected from the co… Show more

“…Novel mathematical treatments of MALDI data have been conceived (Montazery-Kordy et al, 2008;Alexandrov et al, 2009;Liu, 2009). There remains an on-going documentation of blood by MALDI parent ion patterns in response to a variety of physiological states such as disease, drug response, the image or proximity of romantic partners or the use of performance enhancing drugs (Tammen et al, 2002;Guerrier, Lomas, & Boschetti, 2005;Schwegler et al, 2005;de Seny et al, 2005;Orvisky et al, 2006;Ragazzi et al, 2006;Zheng et al, 2006a;Lin et al, 2006b;de Noo et al, 2006b;Au et al, 2007;Cui et al, 2007Cui et al, , 2008bHui et al, 2007;Lim et al, 2007;Taguchi et al, 2007;Tiss et al, 2007;Yildiz et al, 2007;Das et al, 2008;Datta, 2008;Jacot et al, 2008;Kojima et al, 2008;Meuleman et al, 2008Meuleman et al, , 2009Ransohoff et al, 2008;Sundsten, Ostenson, & Bergsten, 2008a;Wei et al, 2008;Zinkin et al, 2008;Han et al, 2008a,b;Liu et al, 2008a;Wang et al, 2008aWang et al, ,c, 2008dCallesen et al, 2008bCallesen et al, , 2009Sundsten et al, 2008b;Albitar et al, 2009;Caron et al, 2009;…”

“…Statistical analysis might be applied to proteomic data at several levels from power calculations and experimental design , peptide patterns (Ragazzi et al, 2006;Ressom et al, 2006;Au et al, 2007;Heidema & Nagelkerke, 2008;Helleman et al, 2008), or parent peptide intensity analysis (Marshall et al, 2003). Statistical test such as t-tests (Negishi et al, 2009), ANOVA (Marshall et al, 2003;Higgs et al, 2008;Zhao et al, 2008a;Sundsten et al, 2008b), goodness of fit tests have also been applied to mass spectrometry of blood Zhu et al, 2006). Peptide fragment patterns might be correlated or identified using a support vector machine (Field, Fenyo, & Beavis, 2002), hypergeometric probabilities (Sadygov & Yates, 2003;Sadygov, Liu, & Yates, 2004), linear modeling Li et al, 2008), cross-correlation analysis (Yates et al, 1995) hidden Markov modeling (Khatun, Hamlett, & Giddings, 2008) or by examining probability distributions (Pappin, Hojrup, & Bleasby, 1993;Perkins et al, 1999;Higdon et al, 2007).…”

“…Based on their m/z , the sample type and fraction in which they were found, the ProteinChip surface used, as well as data from previously performed serum profiling studies (N. Harris, unpublished data, and ), we tentatively identified the most discriminative peaks. The peak with m/z 15,915 that was detected in Fraction 3 on the IMAC30 array is very likely hemoglobin β‐chain.…”

“…The peak with m/z 15,915 that was detected in Fraction 3 on the IMAC30 array is very likely hemoglobin β‐chain. Peaks with similar mass were previously structurally identified as hemoglobin β‐chain by SDS‐PAGE, followed by in‐gel trypsin digestion and analysis using tandem MS (e.g., Q‐TOF), and/or by an immunoassay . The molecular weight (MW) of this protein is 15,867 Da and it has a pI of 6.81.…”

“…SELDI-TOF-MS provides a sensitive and reproducible method to detect and resolve multiple proteins bound to protein chip arrays. The platform has been used in several large-scale proteomic clinical studies on plasma, serum, cerebrospinal fluid and urine [8][9][10][11]. It has further been used as a tool in the development of diagnostic assays of various diseases, including several types of cancer, heart infarction, Alzheimer's disease, rheumatoid arthritis and chronic alcoholism [9,[12][13][14][15][16][17][18].…”
