Plasmonic Fiberoptic Absorbance Biosensor (P-FAB) for Rapid Detection of SARS-CoV-2 Nucleocapsid Protein

Abstract: SARS-CoV-2 nucleocapsid protein-based COVID-19 diagnosis is a promising alternative to the highpriced, time-consuming, and labor-intensive RT-PCR tests. Here, we developed a rapid, dip-type, wash-free plasmonic fiber optic absorbance biosensor (P-FAB) strategy for the point-of-care detection of SARS-CoV-2 N-protein, expressed abundantly during the infection. P-FAB involves a sandwich assay with plasmonic labels on the surface of a U-bent fiber optic sensor probe with a high evanescent wave absorbance (EWA) sens… Show more

“…In addition, preclinical investigations and the development of a PoC read‐out device are in the plans. [ 116 ] A thin coating of GO was formed around the LPFG portion to generate functional groups for the biological recognition element immobilized by covalent bonds, as shown in Figure 3C. [ 117 ] The outside cladding of the LPFG has a greater refractive index than the inner cladding, allowing some cladding modes to be steered in the outer ring‐shaped cladding without interfering with the core mode.…”

“…The proposed U-shaped fiber probe method has excellent sensitivity in the range of parts per billion (ppb), with a response time ranging from 25 to 60 s [30,31]. The steps followed for the fiber-optic-based sensing is summarized step by step in sections below according to the recent work shown in [32,33]. Figure 4…”

“…The gold nanoparticles (AuNp) can be synthesized by citrate-mediated reduction of gold chloride. The biosensor matrix must first be produced by immobilizing gold nanoparticles on a U-bent fiber-optic probe by Satija et al 2014 in [33,34], then covalently conjugating anti-N protein monoclonal antibodies. For this reason, high-affinity antibodies will be obtained.…”

“…In (Singh and Jha 2019 ), fabrication and validation of optical biosensor for non-invasive monitoring of glucose using saliva is proposed where the enzymatic reaction resulted in pH variation which was detected by electronic meter. In (Divagar et al 2021 ), a dip type, wash free plasmonic fiber optic absorbance biosensor (P-FAB) is proposed for point-of-care detection of SARS-CoV-2 N-protein. In (Monfared et al 2021 ), a highly sensitive quasi-D-shaped fiber optic biosensor is proposed for the detection of high refractive index liquid analytes via surface plasmon resonance.…”
